石斛明目胶囊药材水煎醇沉工艺的正交试验 被引量:2

摘要 目的:筛选石斛明目胶囊的水煎醇沉最佳工艺条件。方法:高效液相色谱法测定水煎液和醇沉液中芍药苷的含量;采用正交设计法考察加水量、浸泡时间、煎煮次数、煎煮时间四因素对水煎工艺提取成分提取率的影响;考察醇沉前水煎液相对密度、醇沉浓度、醇沉时间三因素对醇沉工艺有效成分含量的影响。结果:煎煮次数、煎煮时间对水煎工艺有显著影响(P<0.05);醇沉前水煎液相对密度醇沉工艺有显著影响(P<0.05)。结论:结合生产实际,确定石斛明目胶囊水煎最佳工艺是药材不浸泡,第1次煎煮用9倍量水,第2次煎煮用8倍量水,煎煮2次,每次1h;醇沉工艺最佳条件为即煎煮液浓缩密度为1.10~1.15,加入乙醇使含醇量为70%,醇沉时间为12h。 OBJECTIVE To optimize the process of extraction and purification of paeoniflorin by orghogonal design. METH- OIlS The contents of paeoniflorin in water solution were detected by HPLC. Effects of four factors,water volume(A) .extraction time(B), extraction times(C), soaking time(D) on extraction rate were evaluated by orthogonal design. Effects of three factors, concentration of condensed solution(E)), alcohol volume(F), reaction time(G) on content of paeoniflorin of alcohol precipitation were also evaluated by orghogonal design. RESULTS Factor C and D had a significant effect on water-extraction(F〈0. 05) ;Factor E had a significant effect on alcohol-precipitation(F〈0. 05). CONCLUSION The optimum water-extraction factors were A(9 + 8) times, B( 1 ) h, C(2), D(without soak). The optimum alcohol precipitation factors were E( 1.10 - 1.15 g·mL^-1),F(70%),G(12 h).
