摘要
目的研究益脑颗粒对衰老慢性脑缺血大鼠脑组织中血管内皮生长因子(VEGF)的影响,探讨益脑颗粒治疗慢性脑缺血的作用机制。方法采用D半-乳糖(D-gal)注射诱导和永久性双侧大鼠颈总动脉结扎术(2VO)制备亚急性衰老大鼠慢性脑缺血模型。给予高、低剂量益脑颗粒灌胃,并以尼莫地平为对照,免疫组化法观察大鼠脑组织VEGF的表达。结果与正常组比较,模型组脑组织中VEGF表达无明显改变(P>0.05),益脑颗粒高、低剂量组表达升高(P<0.01),尼莫地平组无明显改变(P>0.05)。结论益脑颗粒能增加缺血脑组织中VEGF表达,而增强脑组织血管侧支循环的建立,改善脑缺血。
Objective To observe the effect of Yinao granule on VEGF in brain of chronic cerebral ischemia model of subacute aging rats, and explore the mechanism. Methods D-galactose ( D-gal ) injection and permanent bilateral common carotid artery occlusion (2VO) were used to made model of subacute aging rats. Model rats were feed with high and low-dose Yinao granule, and nimodipine was se- lected as positive control. The expression of VEGF was measured by immuohistochemical method. Results There was no difference of VEGF expression between model group and control group( P 〉0. 05 ) , there were high expressions in high and low-dose Yinao granule groups ( P 〈 0. 01 ), and the high-dose group was superior to low-dose group( P 〈 0. 01 ). There was no obvious change of VEGF expression in nimodipine group. Conclusions Yinao granule can improve the VEGF expression in brain of circulation insufficiency to develop the collateral circulation and play a protective role to perfective cerebral ischemia.
