摘要
目的研究雷公藤多苷(GTW)对雌性幼鼠血清雌激素(E2)含量和卵巢雌激素受体(ER)表达的影响以及对成年后生育力的影响。方法采用4周龄SD雌鼠,据体重随机分为两组,分别为空白对照组、实验组(GTW),连续灌胃12周后,处死部分动物,检测指标。剩余部分大鼠停药,用健康雄性成年SD大鼠按雌∶雄=1∶1合笼2周,继续观察实验雌鼠至分娩后1周。观察受孕率、产仔数、活产率、存活率。结果GTW组的血清E2值高于空白组;ER表达低于空白组,差异有统计学意义(P<0.05)。两组雌鼠的仔数、受孕率、活产率差异没有统计学意义(P>0.05),新产仔存活率组间差异有统计学意义(χ2=10.303,P=0.001)。结论雌性幼鼠服用雷公藤多苷可造成卵巢局部的组织病理改变,但对成年后的生育力无明显影响。
Objective To explore the effect of female impuberal rat's the estrogen in the serum and the expression of the estrogen receptor (ER) in the ovary and uterus caused by multi -glycosides of triptergium wilfordii (GTW) and the procreative damage by GTW. Methods The 4 - weeks age female SD rats were randomly divided into control group and treatment group (GTW) according to weight. All rats were continuously fed with the medicine for 12 weeks, and then cease medication. One was executed, the estrogen were examined by Radi y and the expression of the ER in the ovary and uterus were examined by immuno - histochemistry; The others were put one adult healthy male rat into one cage according the proportion 1 : 1 and being together for 2 weeks. We executed them and explored the effect of the experimental medication on the trial female rat mating, becoming pregnant, gestation and labor course by accounting the pregnant ratio, the amount of the baby rat and their survivor. Results After continuous medication for 12 weeks, the serum estradiol content in GTW group was higher than that in control group and expression of the estrogen receptor in GTW group was lower than that in control group ( P 〈 0.05 ) ; The livability of all groups was 100%. Conclusion This study shows that GTW may damage the configuration of the gonadal tissue, suppressing the expression of the estrogen receptor, but the procreative damage has no obvious effect on the impuberal female rat mating, becoming pregnant, gestation and labor course.
