HIF-1α与COX-2在胃癌中的表达及相关性 被引量:5

摘要 目的探讨缺氧诱导因子-1α(HIF-1α)和环氧合酶-2(COX-2)在胃癌组织中的表达及其相关性。方法对66例手术切除的胃癌标本和15例正常胃黏膜组织进行连续切片,用免疫组织化学方法检测HIF-1α和COX-2蛋白的表达情况。结果胃癌组织中HIF-1α蛋白表达阳性率为68.2%,COX-2蛋白阳性表达率为62.1%,均较正常胃黏膜组织显著增高;COX-2阳性表达与HIF-1α表达呈正相关(r=0.405,P=0.001)。HIF-1α蛋白的表达与胃癌的浸润深度、淋巴结转移及TNM分期密切相关(P<0.05);COX-2蛋白的表达与胃癌的淋巴结转移及TNM分期密切相关(P<0.05)。结论胃癌组织中COX-2和HIF-1α的表达增加与胃癌的不良生物学行为相关,可能成为预测胃癌侵袭转移的重要指标。 Objective To investigate the expression and relationship of HIF -1α protein and COX-2 protein in gastric carcinoma. Methods The immunohistochemical expression of HlF-1α and COX-2 were detected for 66 specimens of gastric carcinoma and 15 cases normal gastric mucosa and the correlation of these expression with clinic opathologic features were examined. Results The protein positive expression rate of HIF-1αlpha and COX-2 in tumor samples were significantly higher than those in normal tissues (P 〈 0.01). The positive expression rate of HIF-lalpha and COX-2 were 68.2% and 62.1% respectively. Expression of HIF-1α protein in gastric carcinoma were associated with their depth of invasion( P=0.044 ),lymphatic node metastasis( P=0.018 ) and TNM stage( P=0.024 ), but not with age, gender, tumor size , degree of tumor differentiation (P〉0.05). The expression of COX-2 protein were associated with their lymphatic node metastasis (P=0.018) and TNM stage(P=-0.042), not with age, gender, tumor size, depth of tumor invasion and degree of tumor differentiation(P〉0.05 ). A positive correlation was found between the expression of HIF-1αand COX-2 protein in gastric carcinoma.( r=-0.405, P=0.001 ). Conclusions The overexpressed of H1F-1α and COX-2 protein are related with poor biological behavior of gastric carcinoma. They can be taken as useful predicators for predicting invasion, metastasis of gastric carcinoma.
