脑梗死急性期气虚血瘀证大鼠神经元凋亡的研究 被引量:2

Neuron apoptosis in rats with acute cerebral infarction of syndrome of qi deficiency and blood stasis

摘要 目的研究脑梗死急性期气虚血瘀证大鼠脑组织神经元凋亡的特征。方法采用持续力竭性游泳复合线栓法建立脑梗死急性期气虚血瘀证大鼠模型,脑组织石蜡切片行脱氧核苷酸末端转移酶介导的核苷酸缺口末端标记法(TUNEL)染色,观察复合模型组与正常组、单纯游泳对照组及单纯线栓对照组大鼠尾状核神经元凋亡的情况,并采用图像分析技术计数凋亡细胞数、分析细胞损伤情况。结果正常组及单纯游泳对照组尾状核仅出现散在凋亡细胞,复合模型组及单纯线栓对照组尾状核出现大量凋亡细胞,复合模型组较单纯线栓对照组凋亡细胞显著增多、细胞损伤更为严重。结论脑梗死急性期气虚血瘀证大鼠脑组织神经元凋亡可以作为微观特征纳入该模型的评价体系。 Objective To compare and study the characteristics of brain neuron apoptosis in rats with acute cerebral infarction of syndrome of qi deficiency and blood stasis. Methods The rat model of acute cerebral infarction of syndrome of qi deficiency and blood stasis was established by using continual exhaustive swimming combined with suture method, and then the brain paraffin sections was stained with Tdt-mediated dUTP nick end labeling (TUNEL). The neuron apoptosis of cerebral caudate nucleus was observed and compared between the combined model group and normal group, single swimming control group or single suture control group. The quantity of apoptotic cells was calculated and cell lesion was analyzed with imaging analysis technique. Results There were a few apoptotic cells in the cerebral caudate nucleus in the normal group and single swimming control group, but a lot in the combined model group and single suture control group. In the combined model group there were much more apoptotic cells and cells were injured more seriously. Conclusion The brain neuron apoptosis, as microscopic characteristics, can be included into the review system of the model in rats with acute cerebral infarction of syndrome of qi deficiency and blood stasis.
