核转录因子-κB在动脉粥样硬化大鼠动脉平滑肌细胞中的表达 被引量:5

摘要 目的探讨核转录因子-κB(NF-κB)在动脉粥样硬化(AS)大鼠动脉平滑肌细胞(SMC)中的表达及AS的发生机制。方法SD雄性大鼠20只随机分为对照组和高脂饮食组,每组10只。对照组用普通饲料喂养,高脂饮食组用高脂饲料喂养。2个月后心脏采血处死大鼠,取胸主动脉,常规固定,包埋,切片。采用免疫组织化学检测各组平滑肌细胞NF-κB p65的表达情况。结果对照组胸主动脉内膜光滑,内皮细胞排列整齐。高脂饮食组动脉内膜脂质浸润,粗糙,隆起,有泡沫细胞形成,细胞排列紊乱。NF-κB p65在对照组动脉SMC呈弱阳性表达,在高脂饮食组SMC胞浆和胞核中有棕褐色颗粒,呈强阳性表达。NF-κB p65在高脂饮食组表达的平均灰度值(86.30±17.29)显著低于对照组(152.36±28.12)(P<0.01)。结论NF-κB参与了AS的发生、发展,高脂血症可能通过诱导血管壁SMC中NF-κB的表达引起AS的发生。 Objective To explore the expression of nuclear factor-kappaB (NF-KB) in arterial smooth muscle cells of rats with atherosclerosis. Methods 20 of male SD rats were randomly divided into control group and high fat diet group, ten rats in each group. The rats of control group were fed with normal diet. The rats of high fat diet group were fed with high fat diet. Two months later, the rats were executed by being taken out the blood from their hearts. The aortas of the rats in each group were incised,fixed,embedded and bladed routinely. The expressions of NF-kB p65 protein in smooth muscle cells were detected by immunohistochemistry. Results The intima of aorta were smooth with only one layer of endothelial cells in control group. The intimas of aorta in the high diet group were crude, hunched and crowded with lipid and foam cells. The expression degree of NF-kB P65 in SMCs of control group was weaker than that in high diet group. There are brown granules located in the cytoplasm and nuclei in high diet group. The value of average grey degree of NF-kB P65 was (86.30 ± 17.29) in high fat diet group,which was evidently lower than that in control group ( 152.36 ± 28. 12 ) ( P 〈 0. 01 ). Conclusion NF-kB may be involved in atherogenesis. Hyperlipemia may lead to atherogenesis by stimulating the expression of NF-kB in SMC.
