摘要
采用连续提取法对苜蓿Medicago sativa样品进行蛋白质分组测定,结果表明:苜蓿蛋白组份随提取剂的种类而变化,蛋白组份含量由大到小依次为40%(v/v)异丙醇>2%(m/v)氯化钠>3.85%(v/v)乳酸>0.5%(m/v)氢氧化钾;其中鲜苜蓿样品中醇溶蛋白占14.50%(m/v);盐溶蛋白占12.55%(m/v);酸溶蛋白占7.73%(m/v);碱溶蛋白占3.11%(m/v),其他蛋白占总粗蛋白的63.11%(m/v)。根据蛋白质组份含量及提取成本确定稀盐溶液为苜蓿叶蛋白适宜的提取剂,盐溶液浓度为0.5%(m/v)。
Adopted the method of continuous extract on determination of grouping protein in alfalfa.The result showed that the alfalfa protein components varied with the type of extract reagents, their contents in order were 40 % (v/v) Isopropanol〉2 % (m/v) Sodium〉3.85 % (v/v) Lactic acid 〉 0. 5 % (m/v) Potassium. Proteins dissolved in alcohol, salt, acid and alkaline accounted for 14. 50%, 12.55%, 7.73%, and 3.11% of the total crude protein, respectively. Others accounted for 63.11%.It was concluded that the most suitable extraction reagent for alfalfa leaf protein was dilute salt solution (0.5%).
