羟丙基-β-环糊精/格列本脲包结物的制备及其表征 被引量:1

Preparation and Characterization of Hydroxypropyl-β-cyclodextrin/Glibenclamide Inclusion Complex

摘要 研究了羟丙基-β-环糊精对格列本脲的包结作用,考察了包结物的性质。通过相溶解度法确定了羟丙基-β-环糊精与格列本脲二者的包结摩尔比和包结稳定常数,采用中和法制备了羟丙基-β-环糊精/格列本脲固体包结物,并通过傅立叶变换红外光谱、差式扫描量热分析、X射线粉末衍射光谱对羟丙基-β-环糊精/格列本脲固体包结物进行了表征。结果表明,格列本脲与羟丙基-β-环糊精的相溶解度图为典型的AL型,两者形成了1∶1的可溶性包结物,在25℃时包结物平衡常数(K1∶1)为303 L.mol-1。羟丙基-β-环糊精/格列本脲包结物的光谱研究结果表明,羟丙基-β-环糊精/格列本脲包结物是格列本脲以无定形状态分散到羟丙基-β-环糊精中。包结物的水溶性实验结果表明包结后格列本脲的溶解度增加了15倍。 Interaction between glibenclamide and hydroxypropyl-β-cyclodextrin was investigated. The stability constant and molar ratio of glibenclamide to hydroxypropyl-β-cyclodextrin was calculated from the phase solubility diagram. The solid inclusion com- plex of hydroxypropyl-β-cyclodextrin/glibenclamide was prepared by neutralization method and characterized by IR, DSC and XRD. It was found that the phase solubility diagram of glibenclamide in HP-β-CD solution showed a typical AL-type, a soluble complex of 1 : 1 molar ratio was formed and the stability constant was 303 L·mol ^-1 at 25℃. The characterization results indicated that in the inclusion complex, glibenclamide was dispersed in hydroxypropyl-β-cyclodextrin in amorphous form. The solubility resuits of inclusion complex in water indicated that the solubility of glibenclamide increased by 15 times.
