摘要
[目的]研究利用SSR技术构建以杂交粳稻亲本为主的SSR指纹图谱。[方法]用48对SSR引物扩增了42个杂交粳稻亲本的基因组DNA,从中筛选出12对具有稳定多态性的引物,推荐为核心引物。建立了42个杂交粳稻亲本×12个核心引物的DNA指纹数据库。[结果]采用类平均法聚类分析计算42个基因型的相似系数,在相似系数0.73处,可将42个杂交粳稻亲本分为5大类。[结论]聚类分析的结果基本反映了品种间的亲缘关系。
[Objective] The study aims to research SSR method establishing the SSR fingerprinting map of parents of japonica hybrid rice.[Method] The genomic DNA of 42 parents of japonica hybird rice is used for PCR analysis with 48 pairs of SSR primers,of which 12 primers are selected as core primers because of their stable polymorphism.A database of SSR fingerprint for 42 genotypes at the 12 primer loci is constructed.[Result] The genetic similarity of 42 genotypes is calculated by cluster analysis of an un-weighted pair-group average method with arithmetic mean,the result shows that 42 varieties of rice can be classified into 5 groups at the level of genetic similarity coefficient is 0.73.[Conclusion]The result of cluster basically reflects the genetic relationships among varieties.
