萱草属部分种和园艺品种的SSR多态性分析 被引量:14

摘要 对萱草14个园艺品种及6个原种或变种在表形性状的基础上辅以SSR分子标记进行聚类分析。在SSR分子标记时,从40对引物中共筛选出多态性引物5对,采用琼脂糖凝胶电泳方法和变性聚丙烯酰胺凝胶电泳方法扩增,多态性百分率分别为91.30%和93.33%;SSR多态性分析后显示,聚类结果分为两类,一类为全部14个供试萱草园艺品种、萱草、黄花菜及萱草的一个变种,此外北黄花菜、小黄花菜和重瓣萱草聚为另一类,这说明实验中所选的萱草园艺品种与萱草H.fulva、黄花菜H.cit-rina的亲缘关系较近。本研究可以为今后有针对性地杂交育种、培育新的萱草园艺品种提供参考。 Analysis of some Hemerocallis species and varietios by SSR-PCR found that five kinds of SSR primers could produce polymorphic bands from forty kinds of SSR primers. The ratios of polymorphie band in agarose and denaturant polyaerylamido gels were 91.30% and 93.33%, respectively. By using five primers in fourteen Hemerocallis horticultural varieties and six Hemerocallis species or varieties, these SSR data were analyzed using cluster methods of UPGMA in the software package NTSYSpe. Twenty materials for trials were divided into two cluster groups. One group was fourteen Hemerocallis varieties for trial, H. fulva, H. citrina, and H. fulva 'Hankow'; H. lilio-asphodelus, H. minor and H. fulva var Kwanso were belonged to another group.It showed these fourteen Hemerocallis varieties have close relationship with H. fulva, H. citrina. This study could provide reference for daylily crossing in future.
