摘要
采用双向凝胶电泳对拟南芥T-DNA插入突变体scc8-D和对照材料cry1cry2的植株总蛋白进行了分离,通过考染显色,获得了分辨率和重复性较好的双向电泳图谱.然后选取30个差异蛋白质点用MALDI-TOF-TOF-MS进行肽质谱指纹图分析,有22个蛋白质点得到了可靠鉴定.其中在scc8-D突变体中有3个蛋白上调,16个蛋白下调,有3个蛋白仅在cry1cry2中表达.分析发现,这些差异蛋白可能参与了碳水化合物代谢、光合作用、光呼吸、胁迫响应、蛋白质合成和氧化还原平衡等过程.这些蛋白中光合蛋白占32%,说明这些与矮化相关的调节性蛋白和功能性蛋白丰度的表达受到光的调控,从而抑制了植物下胚轴的伸长.
To study the difference at the protein expression level of cryl cry2 and T-DNA mutant scc8-D, we used a proteome approach based on 2-DE to compare their protein patterns. 22 differential spots were identified by MALDI-TOF-TOF-MS. In the 2-DE protein patterns of scc8-D, three up-regulated proteins and sixteen down-regulated proteins were identified. Three protein spots only appeared in crylcry2. The identified proteins were involved in several processes, i.e. redox homeostasis, protein synthesis, stress responses, photosynthesis, photorespiration, and metabolisms of carbon. 32 % of the photosynthetic proteins showed degradation in scc8- D. The results suggested that the inhibition of hypocotyl elongation was caused by the expression of regulatory proteins and function proteins, which was under light control.
