摘要
目的:建立高效液相色谱法测定人血浆中万古霉素的药物浓度方法。方法:采用液-液萃取法并选用去甲万古霉素为内标,以Symmetryshield RP18柱为色谱柱。流动相为甲醇:水(以磷酸二氢钾调节pH至3.1)(16:84),流速1.0mL·min^-1。Waters2487双波长紫外检测器,检测波长210nm。结果:线性范围为(0.63-63.0)mg·L^-1,r=0.9996(n=6);平均回收率大于85%,低、中、高浓度样品的日间和日内RSD均小于5%。结论:本法适用于万古霉素的血药浓度测定。
OBJECTIVE To establish an HPLC method for determining plasma level of vancomycin. METHODS The new HPLC procedure involved a liquid-liquid extraction pretreatment procedure and an HPLC internal standard, norvancomycin. A symmetryshield RP18 column was used. The mobile phase of a mixture of methanol water (16:84,pH 3. 1) was used at a flow rate of 1.0 mL·min ^-1. The detector was Waters 2487 Dual λ Absorbance Detector, detector was set at the wavelength of 210 nm. RESULTS The calibration curve was linear in the range of 0. 63 to 63.0 mg· L^- 1. The average recovery was more than 85%, and the RSD within day and between-day were all less than 5%. CONCLUSION This method is suitable for monitoring of vancomycin in plasma.
