摘要
目的:探讨芪参还五胶囊对大鼠脑组织缺血后自体神经干细胞增殖和分化的影响。方法:采用插线法制作大鼠大脑中动脉缺血模型,使用B rdU/NeuN和B rdU/GFAP双重标记和荧光免疫法,分别观察和记录模型组和对照组和芪参还五胶囊治疗组在不同时间点(7、14、21天)上神经干细胞的增殖,并进一步分化成神经元和星形胶质细胞的过程。结果:与模型组和安慰剂对照组相比,芪参还五胶囊治疗组在3个时间点(7、14、21天)上,B rdu+/NeuN+细胞计数和B rdu+/GFAP+细胞计数增多均明显,在统计学上有显著差异(P<0.01)。结论:脑缺血后可诱发内源性神经干细胞发生增殖和分化,经芪参还五胶囊干预可使上述过程明显、持久;推测芪参还五胶囊可能通过增强神经细胞抗缺氧损伤能力、减轻脑组织炎症反应、改善血液黏滞状态、微循环和保护缺血灶周围神经元突触结构的基本完整性等多个环节的共同作用,实现内源性神经干细胞的增殖和分化。
Objective: To discuss the effect of Qishen Huanwu Capsule on the proliferation and differentiation of the endogenous NSC in rats′cerebral ischemia.Methods: The rat models of cerebral ischemia were made into the models of middle cerebral artery occlusion with the intraluminal filament occlusion;double immunofluorescent staining to identify NSC proliferation of the model group and the therapy group at different points(7d,14d,21d),and to differentiate into neurons and astrocytes.Results: BrdU/NeuN and BrdU/GFAP positive cells expression in the therapy group are more than model group,has a significant difference compared with model group(P〈0.01).Conclusion: The cerebral infarction can induce proliferation and differentiation of the endogenous NSC,Qishen Huanwu Capsule therapy can make this process evident and persistent.We confer that Qishen Huanwu Capsule therapy will promote proliferation and differentiation of endogenous NSC through multi-link such as that improvement of the capacity against anoxic injury of neural cell,alleviate inflammatory response of brain,the improvement viscous state of blood,microcirculation,protecting complete neurosynapses around the local ischemia.
