摘要
目的认识不同表面分子标记物所标记的脾脏树突状细胞(dendritic cell,DC)分布、成熟程度与超微结构特点。方法运用光镜、电镜观察与免疫组化标记(CD11c、CD205、CD209、S-100、CD1a、HLA-DR、CD80及CD86)方法观察35例正常人脾脏组织中的树突状细胞的形态分布及超微结构特点。结果正常人脾脏树突状细胞主要分布在白髓边缘区和T细胞区,CD11c、CD205和CD209是目前较特异的树突状细胞标志物,标记阳性率分别为4.19%、3.96%和3.02%,其中CD209+ DC分布更靠近红髓。树突状细胞表达共刺激分子CD80与CD86的阳性率分别为2.68%和2.02%,主要分布在T细胞区。树突状细胞超微结构特点为细胞体积较大,呈细长状或不规则形,表面可见数个突起向周围细胞间隙延伸;胞核形状不规则,表面有凹陷,可见核仁;胞浆致密或呈低电子密度,细胞器欠发达。结论正常人脾脏中不同表面分子阳性树突状细胞的比率和分布有所不同,其中大部分为不成熟的树突状细胞,具有活性的成熟树突状细胞数目较少。
Objective To observe the distribution of human splenic dendritic cells (DCs) with several DC markers and uhramicrostruetural features of these DCs. Methods Spleen sample tissues of 35 individuals, including 30 patients after acute traumatic splenic rupture and 5 corpses after sudden death due to cardiac or cerebrovascular diseases were collected. The splenic dendritic cells in these sample sections were observed and analyzed by light microscopy, electron microscopy, and immunohistochemical staining with DCs markers including CDlle, CD205, CD209, S-100, CDla, HLA-DR, CD80 and CD86. Results DCs were mainly distributed in 3 distinct regions, the T-cell zones, the B-cell zones, and the marginal zone in the spleen. In the marginal zone, DCs formed a ring surrounding the white pulps. CD209 + DCs were nearer to the red pulp than CD11 c * DCs and CD205+ DCs. The CD11 c + DCs, CD205 + DCs and CD209 + DCs accounted for about 4.19% , 3.96% and 3.02% of all spleen mononuelear cells respectively. In normal human spleen, most DCs were immature and only a small amount were mature DCs, which were CD80 or CD86 positive and mainly distributed in the T-cells zones. Under an electron microscope, splenic DCs were large, slender or irregular in cell body, with several protrudes extending into the intercellular space. The nuclei were irregular in shape and depressed in surface with clear nucleoli. The cytoplasm was compacted or in low electron density and some organelles were seen. Conclusion Splenic DCs with different DC markers locate in different regions of the spleen. Most of them are immature, and only a small amount is mature.
