超声辐照载羟喜树碱微泡对肝癌SMMC-7721细胞增殖的抑制作用

摘要 目的探讨超声辐照载羟喜树碱微泡对肝癌细胞株SMMC-7721的增殖抑制作用。方法将SMMC-7721细胞随机分为6组,即载羟喜树碱微泡+超声组、羟喜树碱+超声组、单纯羟喜树碱组、载羟喜树碱微泡组、空白微泡+超声组及对照组。采用MTT法观察各处理因素对细胞的生长抑制情况;流式细胞仪(FCM)分析细胞周期分布;Annexin V-FITC/PI双染法和TUNEL法检测细胞凋亡状况。结果载羟喜树碱微泡+超声组对肝癌细胞株SMMC-7721的增殖有明显抑制作用,其24h、48h、72h的抑制率分别为(49.56±1.57)%,(80.01±1.52)%,(91.41±0.64)%;FCM结果显示其将SMMC-7721细胞有效阻滞于G0/G1期;Annexin V-FITC/PI检测细胞早期凋亡率为(12.18±1.38)%;TUNEL法显示染成棕黄色的凋亡细胞,细胞中晚期凋亡率为(34.25±1.83)%。以上结果与其他组的检测结果相比,差异有统计学意义。结论超声辐照载羟喜树碱微泡能明显抑制肝癌SMMC-7721细胞的增殖。其机制可能与阻滞细胞周期于G0/G1期同时诱导细胞凋亡有关。 Objective To investigate the proliferation inhibition of hydroxycamptothecin (HCPT)-carrying microbubbles on human hepatoma SMMC-7721 cells using technique of ultrasound mediated drug release. Methods The SMMC-7721 cells were randomly divided into six groups as follows: HCPT-carrying microbubbles plus ultrasound, HCPT plus ultrasound, HCPT, HCPT-carrying microbubbles, blank microbubbles plus ultrasound, and control group. The cell growth inhibitory effect of each group was observed by MTT assay. Cell cycle distribution was analyzed using flow cytomertry (FCM). Apoptosis was ascertained by Annexin V-Fitc/PI and TUNEL methods. Results The growth of SMMC-7721 cell line was significantly inhibite by combining HCPT-carrying microbubbles and ultrasound. The cell inhibitory rate at 12 h, 48 h, 72 h was (49.56±1.57)%, (80.01±1.52)% and (91.41±0.64)%, respectively. The SMMC-7721 cells were effectively arrested in G0/G1 phase. The apoptosis rate at early term was found to reach (12. 18±1.38)% by Annexin V-Fitc/PI assay. Browncolored positive apoptotic cells were observed and the apoptosis rate at advanced term was found to reach (34.25±1.83) with TUNEL method. Differences with other groups were significant. Conclusion The method of ultrasound irradiating HCPT carrying microbubbles significantly inhibits the human hepatoma SMMC-7721 cells proliferation. Cell cycle arrest and apoptosis might be responsible for cells growth inhibition.
