农杆菌介导茎尖转化体系对不同基因型棉花转化率的影响 被引量:10

摘要 选用两种不同基因型陆地棉华抗6号和06S62为材料,以无菌黄化苗茎尖为受体,用农杆菌介导法进行转化,对不同的菌液浓度、浸染时间和负压处理等进行了比较试验,探讨农杆菌介导对两种不同基因型棉花遗传转化率的影响。结果表明,农杆菌介导棉花转化的适宜条件是:对于华抗6号,菌液浓度OD600达到0.7、浸染时间10min、负压处理10min时,转化效果较好;对于06S62,菌液浓度OD600达到0.9、浸染时间15min、负压处理15min时,转化效果较好;华抗6号的转化率高于06S62,其最高转化率为8.81%。通过该茎尖遗传转化体系,将bar导入上述两种基因型棉花获得了抗除草剂的转基因植株。 The effect of Agrobacterium-mediated on transformation frequency of different genotypes of cotton was studied in this experiment. Huakang6 and 06S62 were selected as the materials, which were two different genotypes of upland cotton. By changing the different of concentration of Agrobacterium, duration of infection and negative pressure, the proper transformation condition was confirmed. The results showed that the suitable conditions of Agrobacterium-mediated transformation of cotton was that Huakang6, the concentration of Agrobacterium of OD600 0.7, duration of infection for 10min, negative pressure for 10rain was better. 06S62, the concentration of Agrobacterium of OD600 0.9, duration of infection for 10min, negative pressure for 10rain was better. The transformation frequency of Huakang6 was higher than that of 06S62. Its peak transformation frequency was 8.81%. Using the genetic transformation system, bar was introduced into the two genotypes of cotton, and genetically modified herbicide-resistant plants were obtained.
