摘要
目的:通过对过山枫乙醇提取物对大鼠胶原诱导性关节炎模型的实验研究,探讨过山枫治疗类风湿性关节炎的药理作用机制和进行安全性评估,为下一步新药开发研究提供依据。方法:制作类风湿性关节炎大鼠模型,肾功能影响采用脲酶-BER ttelo法。结果:醇提小剂量组血清SOD含量显著高于模型组。给药30天,GSF对Ⅱ型胶原诱发关节炎模型大鼠肝脏功能无明显毒副作用,模型大鼠谷丙转氨酶、谷草转氨酶活性显著高于对照组。给药5周后,胸腺指数均高于模型对照组,水提组与模型组比较差异显著;实验组肝脏指数小于模型对照组。结论:过山枫乙醇提取物具有抗炎作用;过山枫乙醇提取物在治疗浓度时,对肝脏、肾脏和子宫等重要脏器没有明显的毒副作用。
Objective:With the purpose of providing lab groundwork data and lay a foundation for the future research,development and clinical applications of GSF′s ethanol extract,we established animal models of RA,and observed the therapeutic effects of GSF′s ethanol extract on RA,and research the pharmacology effects and the security of GSF′s ethanol extract.Methods:Choose Wistar rats(SPF,130~170g)to make animal model of RA provided by the Animal Centre of the First Military Medical University.the pharmacologicaly effects were detected with the function of anti-oxygenation,anti-inflammation and adjust immunity,and the security of GSF with the weights of rat,ALT,AST,BUN,Cr and the exchanges of viscera.Animal model was divided into five groups at random: control group(administered with NS),Tripterygium Glucosides group(administered with Tripterygium lucosides),GSF high-dosage group(42g/kg),GSF mid-dosage group(28g/kg),GSF low-dosage group(14g/kg).Observed the degree of red and swollen of the back leg of rats;content of SOD in blood serum.Observed the Security of GSF′s ethanol extract,observed the liver function by testing ALT and AST,and the kniy function by testing BUN and Cr;also observed the weight of rats,and the quotiety of viscera include thymus,spleen,liver,womb and ovary.Results:There was no distinct difference between control group and model control group of SOD in blood serum,but GSF low-dosage group was much higher than in model control group(P〈0.05).GSF can control the inflammation of immunity arthritic with type Ⅱcollagen,especially in GSF high-dosage group.There was no oxicity and side-effects to liver.No toxicity and side effects to kidney.BUN and Cr belong to normal range.No difference among groups in two weeks;and at the third week,the weight of rats in GSF and Tripterygium Glucosides group was higher than those in model control group;and in the forth and fifth week,the weight of rats in all groups is go up quickly.It indicated that there is not evident infecti
