应用C57BL/6小鼠进行丙烯酰胺的感觉和运动行为毒性的研究 被引量:2

Study on Toxicity of Sensory and Motor Behavior on C57BL/6 Mice with Acrylamide

摘要 目的用丙烯酰胺造成C57BL/6小鼠的感觉和运动行为损伤,应用OECD推荐的感觉和运动行为测试方法测试,以获得稳定而敏感的感觉-运动行为测试组合。方法选用雄性C57BL/6小鼠分别以5、10、20、30mg/kg BW剂量的丙烯酰胺灌胃,对照组给予蒸馏水,每周5次,连续3周,每周测量小鼠体重、热觉传导、转棒、抓力、后肢撑力等指标。结果染毒后小鼠体重与对照组相比差异无统计学意义(P>0.05),热觉传导加快(10mg/kgBW剂量组在染毒2周后小鼠舔后足潜伏时间缩短了26%,5mg/kgBW剂量组在染毒3周后潜伏时间缩短28%)。在转捧上停留的时间缩短(30mg/kg.BW剂量组小鼠染毒2周后在转棒上的停留时间缩短了19%,染毒3周后缩短了42%),抓力下降(20、30mg/kgBW两个剂量组在染毒3周后抓力分别降低了19%和29%),后肢展开距离明显加宽(10、20、30mg/kgBW小鼠后肢展开距离均加宽),与对照组相比,均有统计学意义(P<0.05)。结论C57BL/6小鼠适合于感觉-运动行为毒性的研究,应用热板试验、转棒试验、抓力试验、后肢支撑力试验组合可以初步筛查感觉-运动神经损伤毒物。 Objective To cause dysfunction of sensory and motor behavior of C56BL/6 mice with acrylamide and to use the method of sensory and motor test that was recommended by Organization for Economic Cooperation and Development (OECD) to test in order to obtain a stable and sensitive test combination of sensory-motor function. Method Seventy five C57BL/6 male mice were randomly divided into 5 groups with 15 mice in each group. The mice in different treatment groups received distilled water (control group), 5, 10, 20 and 30 mg/kg BW of acrylamide for 3 weeks (5 days per week) by gavage. Neurobehavioral indexes including body weight, hotplate, rotarod, grip strength, hind limb landing foot splay were determined every week. Results The body weights of mice treated with acrylamide were not statistically different from that in the control group (P 〉 0.05). The conduction of sensory nerve to heat increased (The latency time of licking hindfeet of mice in 10 mg/kg BW dose group after 2 weeks deceased 26%, and the latency time in 5 mg/kg BW dose group after 3 weeks deceased 28% ). The time staying on the rotating cylinder decreased (After 2 weeks, the time in 30 mg/kg BW dose group decreased 19%, and after 3 weeks decreased 42% ). The grip strength deceased (After 3 weeks, the grip strength in 20 and 30 mg/kg BW dose group decreased 19% and 29%, respectively). The hind limb landing foot splay increased (The hind limb landing foot splay in 10 mg/kg, 20 mg/kg, 30 mg/kg BW dose group all increased). Those indexes in each treatment group were statistically different from that in the control group (P 〈 0.05). In comparison with the previous literature, C57BL/6 mice on the toxicity of sensory and motor behavior were more sensitive. Conclusion C57BL6 mice could be suitable to study toxicity of sensory and motor function. The tests of hotplate, rotarod, grip strength, measurements of hind limb landing foot splay could be screen the toxicants of sensory and motor nerve dysfunction.
