摘要
为了解大鼠肝肿瘤成模过程中肝组织结构变化,用二乙基亚硝胺(diethylnitrosamine,DENA)灌胃法构建大鼠肝肿瘤模型,按常规方法制备肝组织石蜡切片和镜检.建模第1周大鼠肝脏的肝小叶结构完整,肝细胞排列整齐.从第5周起肝窦扩张,肝细胞变性肿胀并有不同程度的脂肪变性,同时出现炎症细胞浸润.第8周时肝窦充血,肝细胞脂肪变性严重,并出现纤维组织增生和明显的肝细胞增生.第10周时肝小叶结构受到破坏,出现肝细胞增生性结节.第12周时门管区内有不同程度的卵圆细胞增生及结缔组织增生,伴随胆管增生.第16周时肝小叶结构消失,可见典型的假小叶结构,结节内肝细胞异常增生明显,出现"结中结",病理性核分裂相亦较多.第17周时肝肿瘤形成,且主要是肝细胞癌,癌细胞排列为索状、腺状、实片状.根据上述结果得出如下结论:用二乙基亚硝胺灌胃法构建大鼠肝肿瘤模型方便有效,肝组织结构和病理变化有规律,易于观察分析,值得推广应用.
To explore systematically the changes of hepatic histology in the progression of cancer liver in rats,the rat liver cancer was induced by perfusing diethylnitrosamine gastric.The hepatic tissue was embedded in paraffin wax and sliced according to the routine method,and then its histology was observed by a microscope.Rat hepatic lobule structure was intact,and liver cells were arranged regularly in the first modeling week.Sinusoid from the fifth week of expansion,swelling and degeneration of liver cells has varying degrees of fatty degeneration,infiltration of inflammatory cells at the same time.In the eighth week sinusoidal congestion,fatty degeneration of liver cells is a serious one and appeared fibrous tissue hyperplasia and liver cell proliferation significantly.In the tenth week,hepatic lobule structure is subject to damage in companying with nodular hyperplasia of liver cells.In the twelfth week,portal area has various degrees of oval cell hyperplasia and connective tissue hyperplasia,accompanied by ductular proliferation.In the sixteenth week,hepatic lobule structure disappeared,showing a typical pseudo-lobular structure and obvious nodular liver cell dysplasia,a"knot in the knot",pathological mitotic phase.Liver tumor formation is successful in the seventeenth week and is mainly in form of hepatocellular carcinoma,cancer cell ranked as the cord-like,gland-like or flaky.The results mentioned demonstrated that rat model of cancer liver induced by perfusing diethylnitrosamine gastric was economical and convenient.The changes of hepatic histology and pathology were so regular and easy to be observed and analyzed that this rat model of cancer liver is worthy to be popularized and applied.
