摘要
[目的]研究北五味子的组织培养,为其快速繁殖提供参考。[方法]以北五味子的嫩茎段、叶片、叶柄为外殖体,MS和改良B5为基本培养基,使用不同激素不同浓度配比,优选不同外殖体诱导愈伤组织的最佳培养基。[结果]嫩茎段的最佳诱导培养基为:改良B5+6-BA 2.5 mg/L+NAA 0.1 mg/L;叶片的最佳诱导培养基为:改良B5+6-BA 2.0 mg/L+NAA 0.1 mg/L+2,4-D 1.0 mg/L;叶柄的最佳诱导培养基为:改良B5+6-BA 2.5 mg/L+2,4-D 1.0 mg/L。7种培养基均未诱导出根。[结论]改良B5培养基优于MS培养基,嫩茎段为愈伤组织诱导的最佳外殖体。
[Objective]The aim was to study the tissue culture of Schisandra chinensis and provide preference for papid propagation.[Method]The tender stem segments,leaves and petioles of Schisandra chinensis were cultured as explants with various concention media(MS and improvement B5) and different kinds of plant growth regulator.[Result]The result showed that improvement B5 medium supplemented with 2.5 mg/L 6-BA and 0.1 mg/L NAA had the best effect on inducement of the tender stem segments,improvement B5 medium supplemented with 2.0 mg/L 6-BA, 0.1 mg/L NAA and 1.0 mg/L 2,4- D had the best effect on leaves inducement and improveraont B5 medium supplemented with 2.5 mg/L 6-BA and 1.0 mg/L 2,4-D had the best effect on petioles inducement . Seven kinds of midia didn't induce root. [ Conclusion]Improvement B5 edium was better than MS medium, the tender stem segment was good explant of callus induction.
