本文比较了头孢呋肟(cefuroxime),头孢氨噻肟(cefotaxime) 和头孢氧哌唑(cefoperazone)对7种标准β-内酰胺酶及6株临床分离的产酶耐药菌株的酶提取物的稳定性,实验采用紫外分光光度法.实验结果表明,头孢呋肟的酶稳定性与头孢氨噻肝相似,优于头孢氧哌唑.对头孢呋肟和头孢氨噻肟的正常人体药代动力学研究表明,头孢呋肟与头孢氨噻肟的人体药代动力学特性亦基本和似.药物的血尿药物浓度测定采用微生物法.
The beta-lactarmase stability and the human pharmacokinetics ofcefuroxime were investigated and compared with cefopeazone and cefotraxime, both ofwhich are third generation cephalosporins. The enzyme stability of cefuroxime and theother two cephalosporins towards 7 standard beta-lactamases and cell-free enzyme ex-tracts from 6 clincal isolates was measured using Hitachi 557 double - wavelengthdouble-beam spectrophotometer. Single 1.0 g intravenous doses or cefuroxime andcefotaxime were given to six volunteers separately in the crossover design study with a timeinterval of 2 weeks. Concentrations of the antibiotics in specimens of the antibiotics inspecimens er serum and urine were deter-mined by microbiological assay. The beta-lactamase stability of cefuroxime was very similar to that of cefotaxime andmuch better than that of cefoperazone. The favorable pharmacokinetic properties ofcefuroxime in human were also very similar to those of cefotaxime .
