摘要
目的:观察和营安心方对慢性心衰大鼠心功能及心肌细胞内钙离子浓度的影响。方法:采用皮下注射较大量异丙肾上腺素(ISO)的方法制备慢性心衰大鼠模型,随机分为模型对照组、卡托普利组、参麦注射液组、和营安心方大、中、小剂量组,另设正常对照组,每组9只。测定左室内压峰值(LVSP)、左室舒张末压(LVEDP)、左室内压上升、下降最大速率(±dp/dtmax)和心重指数,用流式细胞仪测定心肌细胞内钙离子浓度。结果:模型对照组心重指数增加,LVSP、±dp/dtmax降低,LVEDP升高,心肌细胞内钙离子浓度明显升高,与正常对照组比较,差异有统计学意义(P<0.01)。和营安心方大、中剂量治疗组能降低心重指数,改善心功能,降低心肌细胞内钙离子浓度,与模型对照组比较,差异有统计学意义(P<0.05或P<0.01)。结论:和营安心方对慢性心衰大鼠心肌细胞内钙超载、心功能有良好改善作用。
Objective： To study the e ects of Heying Anxin Fang on cardiac function and intracellular Ca^2＋ concentration in cadiocyte of chronic congestive heart failure rats. Methods： Animal models of heart failure were induced by subcutaneous injection of a large dose of ISO, and were randomly divided into model group, Shenmai injection group, captopril group, high, middle and low dosages of Heyinganxin Fang group and normal group with 9 in each group. To determine the indexes of LVSP, LVEDP, ＋dp/dtmax ,-dp/dtmax and heart weight, and determine Ca^2＋ concentration in cardiomyocytes by ow cytometer. Results： Compared with normal group, the indexes of heart weight, Ca^2＋ concentration and LVEDP in model group increased, but indexes of LVSP, ＋dp/dtmax, -dp/dtmax decreased obviously（P〈0.01）. Compared with model group, high dosage and middle dosage of Heyinganxin can decrease Ca^2＋ concentration, reduce heart weight index and improve cardiac function（P〈0.05 or P〈0.01）. Conclusion： Heyinganxin Fang can inhibit intracellular calcium overload in cadiocyte and improve cardiac function of chronic congestive heart failure, it is an e ective drug in treating congestive heart failure.
