抑制性杀伤细胞受体在NK细胞和T细胞上的表达及其与急性移植物抗宿主病的关系

The expression of natural killer cells inhibitory receptors on natural killer cells and T lymphocytes and the relationship with acute graft-versus-host disease

摘要 目的研究异基因造血干细胞移植(allo-HSCT)患者移植前后外周血NK及T细胞上4种抑制性杀伤细胞受体(CD_(158a)、CD_(158b)、NKB1和CD_(94)/NKG2A)的表达及其与急性移植物抗宿主病(aGVHD)的关系。方法采用流式细胞术检测NK及T细胞上抑制性杀伤细胞受体的表达。结果NK细胞上CD_(158a)和CD_(158b)的表达于移植后3~4个月、NKB1和CD_(94)/NKG2A的表达于移植后2个月恢复移植前水平。移植前后CD_(158a)和NKB1在CD_3^+T细胞上持续低水平表达;移植前CD_(158b)和CD_(94)/NKG2A在CD_3^+T细胞上的表达水平较高,且主要表达于CD_8^+T细胞上,移植后其在CD_8^+T细胞上的表达增加。CD_8^+T细胞上CD_(158b)的表达在发生Ⅰ度aGVHD时显著增高,与无aGVHD组及Ⅱ~Ⅳ度aGVHD组相比,差异均有统计学意义(P均<0.05);在发生Ⅱ~Ⅳ度aGVHD时增高不明显,与无aGVHD组相比,差异无统计学意义(P>0.05)。结论CD_(158b)在CD_8^+T细胞上高表达可能有助于降低T细胞同种反应性,减轻aGVHD的程度。 Objective To investigate the expression of four type natural killer(NK) cells inhibitory re- ceptors (CD_(158a),CD_(158b),NKB1 and CD_(94)/NKG2A) on peripheral blood NK cells and T lymphocytes in patients before and after allogeneic hematopoietic stem cell transplantation (allo-HSCT) and the relationship with acute graft-versus-host disease(aGVHD).Methods Flow cytometry analyzes the expression of four type NK cells inhibitory receptors on NK cells and T cells.Results The expression of CD_(158a) and CD_(158b) on NK cells was recovered 3~4 months after allo-HSCT,while NKB1 and CD_(94)/NKG2A was recovered 2 months after allo- HSCT.The percentage of CD_(158a) and NKB1 expressing on CD_3 positive T cells was low and there was little change after allo-HSCT.The percentage of CD_(158b) and CD_(94)/NKG2A expressing on CD_3 positive T cells be- fore transplantation was relatively higher compared with other NK cells inhibitory receptors,such as CD_(158a) and NKB1,and mainly on CDs positive T cells.The expression of CD_(158b) and NKG2A on CD_8 positive T cells was increased after allo-HSCT.The percentage of CD_(158b) expressing on CD_8 positive T cells was significantly in- creased in patients with gradeⅠaGVHD compared with those without aGVHD and those with gradeⅡ~ⅣaGVHD (P<0.05,respectively),while no significantly increased in patients with gradeⅡ~ⅣaGVHD com- pared with no aGVHD (P>0.05 ).Conclusion The high expression of CD_(158b) on CD_8 positive T cells may fa- vor to control T cells alloreactivity and reduce aGVHD.
