摘要 目的通过研究肿瘤患者外周血自然杀伤(NK)细胞表面NKG2A和NKG2D的表达情况,探讨二者表达失衡与肿瘤免疫逃逸的关系。方法采用流式细胞术检测40例肿瘤患者和10名正常对照者外周血NK细胞表面NKG2A和NKG2D的表达情况。用全自动五分类血液分析仪测定各病例的血常规。结果肿瘤患者和正常对照组白细胞数值差异无统计学意义;肿瘤患者淋巴细胞比率低于对照组,而NK细胞明显高于对照组;肿瘤患者NK细胞表面抑制性受体NKG2A和活化性受体NKG2D均明显高于对照组,但肿瘤患者NKG2D荧光强度呈降低趋势。结论肿瘤患者NK细胞表面NKG2A/NKG2D表达失衡,可能是肿瘤免疫逃逸的机制之一。 Objective To investigate the expression of NKG2A and NKG2D on peripheral nature killer(NK) cell of patient with tumor and discuss the relation between imbalance expression of NKG2A and NKG2D on peripheral NK cell and tumor immune escape. Methods The expression of NKG2A and NKG2D on peripheral NK cells was detected in 40 patients with tumor and 10 healthy controls by cytometric analysis. The level of peripheral blood cells in every case was detected by automated hematology analyzer with five classifications. Results The level of peripheral white blood cell of patient with tumor was similar to healthy controls ; While the level of peripheral lymphocyte decreased and the level of peripheral NK cell significantly increased compared to healthy controls; The expression of inhibitory receptor NKG2A and activating receptor NKG2D on peripheral NK cell significantly increased respectively and fluorescent intensity of NKG2D tended to decrease compared to healthy controls. Conclusions Imbalance expression of NKG2A and NKG2D on peripheral NK cell of patient with tumor may be an important factor in tumor immunological escape.
