摘要
目的观察南蛇藤乙酸乙酯、正丁醇提取物联合抗癌化疗药物对体外培养的人SGC-7901、HeLa细胞增殖的影响。方法采用MTT法检测不同浓度的南蛇藤乙酸乙酯、正丁醇提取物(15、30、60、120μg/ml)对SGC-7901、HeLa细胞增殖的抑制率,计算出各自的IC50浓度。用南蛇藤乙酸乙酯、正丁醇提取物IC50浓度分别与紫杉醇5.0μg/ml、卡铂40.0μg/ml、环磷酰胺40.0μg/ml、表阿霉素0.4μg/ml、顺铂3.0μg/ml、5-氟尿嘧啶10.0μg/ml单用、联合使用24 h,检测出各组的抑制率。利用金正均法评价南蛇藤提取物与常用抗癌药物联用的效果。结果不同浓度的南蛇藤提取物对HeLa细胞均有良好的抑制作用,呈剂量依赖性关系(P<0.01)。南蛇藤乙酸乙酯、正丁醇提取物IC50浓度分别72.13μg/ml(SGC-7901)、325.62μg/ml(HeLa);79.36μg/ml(SGC-7901)、355.31μg/ml(HeLa)。各常用抗癌药物对SGC-7901、HeLa细胞增殖的抑制率,紫杉醇、卡铂、环磷酰胺、表阿霉素、顺铂、5-氟尿嘧啶分别为(38.69±0.74)%、(21.91±1.87)%、(35.46±1.31)%、(36.49±3.00)%、(35.84±3.51)%、(36.33±1.60)%;(36.35±0.75)%、(18.65±1.08)%、(29.59±1.64)%、(34.51±3.21)%、(33.17±2.52)%、(21.32±1.33)%。南蛇藤乙酸乙酯、正丁醇提取物IC50浓度与抗癌药物联合使用抑制率均高于单用,且具有协同或相加作用。结论南蛇藤乙酸乙酯、正丁醇提取物与抗癌药物联合使用,具有协同或相加作用,可作为抗癌药物的增敏剂。
Objective To observe proliferation effects of ethyl acetate extract and n-butanol extract of Celastrus orbiculatus Thunb combined with chemotherapeutic agents on SGC-7901 and HeLa cell in vitro.Methods SGC-7901 and HeLa cells were incubated with different doses of ethyl acetate extract and n-butanol extract of Celastrus orbiculatus Thunb alone or combined with Tax(5.0 μg/ml),CBP(40.0 μg/ml),CTX(40.0 μg/ml),E-ADM(0.4 μg/ml),DDP(3.0 μg/ml)and 5-Fu(10.0 μg/ml)for 24 hours,respectively.The inhibition ratio and IC50 were calculated by MTT assay.In addition,the combination effect between the Celastrus orbiculatus Thunb effective constituents and chemotherapeutic agents was detected with Professor Jin′s evaluation methods.Results The proliferation of SGC-7901 and HeLa cells was obviously inhibited by the extracts of Celastrus orbiculatus Thunb(P<0.01).The IC50 of ethyl acetate extract and n-butanol extract of Celastrus orbiculatus Thunb were 72.13 μg/ml and 79.36 μg/ml in SGC-7901 cells;325.62 μg/ml and 355.31 μg/ml in HeLa cells,respectively.The inhibition ratio of SGC-7901 treated with Tax,CBP,CTX,E-ADM,DDP and 5-Fu was(38.69±0.74)%,(21.91±1.87)%,(35.46±1.31)%,(36.49±3.00)%,(35.84±3.51)% and(36.33±1.60)%,respectively.The similar effect on HeLa cells was(36.35±0.75)%,(18.65±1.08)%,(29.59±1.64)%,(34.51±3.21)%,(33.17±2.52)%,(21.32±1.33)%,respectively.The combination of the extract of Celastrus orbiculatus Thunb and chemotherapeutic agents showed a synergetic or additive role in inhibiting cell proliferation,compared with cells treated alone(P<0.01).Conclusions The extracts of the Celastrus orbiculatus Thunb could be used as a chemotherapeutic sensitizer in clinic.
