双氢睾酮与比鲁卡胺对前列腺癌LNCaP细胞系HMGN5基因和AR基因转录和表达的影响 被引量:2

Effect of dihydrotestosterone on gene transcriptions and expressions of HMGN5 and AR in LNCaP cell line

摘要 目的探讨双氢睾酮(DHT)对前列腺癌LNCaP细胞系HMGN5基因和AR基因转录和表达的影响。方法用添加10%碳吸附胎牛血清的RPMI-1640(无酚红)培养基培养LNCaP细胞系24h后,加入不同浓度DHT(0、0.1、1、10、50、100nmol/L)和比卡鲁胺(Casodex)(20μmol/L)。定量反转录qRT-PCR检测LNCaP细胞中HMGN5和AR mRNA水平,免疫印迹法检测HMGN5和AR蛋白表达情况。结果用不同浓度DHT和Casodex培养激素依赖性前列腺LNCaP细胞系24h后,随着DHT浓度升高,LNCaP细胞中HMGN5和AR mRNA水平表达增高,组间差异有统计学意义。AR蛋白表达增高,HMGN5蛋白表达无明显差异。结论 DHT增强了雄激素依赖性前列腺癌LNCaP细胞系中HMGN5 mRNA的表达,这种作用可以被比卡鲁胺减弱。 Objective To evaluate effect of dihydrotestosterone(DHT) on gene transcriptions and expressions of HMGN5 and AR in prostate cancer of LNCaP cell line.Methods The human prostate cancer of LNCaP cell line was maintained in RPMI-1640(no phenol red) supplemented with 10% Carbon adsorption of fetal bovine serum.24 hours later,LNCaP was treated with different concentrations of DHT(0,0.1,1,10,50,100 nmol /L) and Casodex(20 μmol /L) in RPMI-1640(no phenol red) medium.The gene transcriptions and expressions of HMGN5 and AR were examined by qRT-PCR and Western Blot.Results Transcription of HMGN5 and AR mRNA was increased compared with control group which was treated without any DHT or Casodex 24 hours later(P < 0.05).This trend can be suppressed when medium was supplemented Casodex.However,only the expression of AR protein was enhanced.Conclusions DHT can enhance the transcription of HMGN5 in human prostate cancer LNCaP cell line,which can be antagonized by Casodex.
