摘要
背景:骨髓间充质干细胞成骨分化与成脂分化方向对骨再生和修复的影响是目前的研究热点,过氧化物酶体增殖物激活受体γ特别是过氧化物酶体增殖物激活受体γ2和核心结合因子α1对骨髓间充质干细胞分化方向有调控作用。目的:分析2型糖尿病大鼠骨髓腔内过氧化物酶体增殖物激活受体γ和核心结合因子α1的表达变化与骨折愈合障碍的关系。设计、时间及地点:随机对照动物实验,于2007-08/12在中南大学第三附属医院中心实验室完成。材料:选用雄性SD大鼠30只,按随机数字表法分为2组,对照组10只,实验组20只,分别给予普通饲料、高糖高脂饲料喂养。方法:大鼠喂养8周后断尾法取血,实验组腹腔注射链脲佐菌素30mg/kg,对照组注射等量枸橼酸盐缓冲液。注射2周后断尾法取血,建立大鼠左胫骨牵引成骨模型,行左胫骨中上段骨干横行截骨,安置特制延长外固定架,牵引14d。主要观察指标:行X射线摄片比较2组大鼠牵引间隙内骨痂的形成;在组织学水平观察牵引骨痂内微骨柱及初始基质前沿形成及骨折两端髓腔内脂肪细胞形成,并计算骨折两端髓腔内脂肪细胞与骨髓腔面积百分比;以反转录-聚合酶链反应法检测双侧股骨骨髓腔内过氧化物酶体增殖物激活受体γ和核心结合因子α1mRNA的表达。结果:实验组大鼠高糖高脂饲料喂养8周后,总胆固醇、三酰甘油及空腹胰岛素浓度均高于对照组(P<0.05~0.01);实验组大鼠腹腔注射链脲佐菌素2周后:空腹血糖、总胆固醇及三酰甘油浓度高于对照组(P<0.05~0.01),说明2型糖尿病建立成功。X射线摄片显示,实验组大鼠骨折断端之间牵引骨痂形成明显减少,骨痂组织学检查表现为微骨柱基质排列紊乱,初始基质前沿浅染。组织学检查显示,实验组大鼠骨折两端髓腔内脂肪细胞及脂肪细胞占骨髓腔面积百分比高于对照组(P<0.01)。反转录-聚合酶链�
BACKGROUND: The osteogenic differentiation and adipogenic differentiation of bone marrow mesenchymal stem cells (MSCs) is the hot topic in recent research. Peroxisome proliferators activated receptor γ (PPAR γ ), especially PPAR γ 2 and core binding factor α 1 (CBF α1) has regulation effect on the differentiation of MSCs. OBJECTIVE: To investigate the relationship between impaired fractures healing and the expression of PPAR γ and CBF α1 in fracture union disorder of rats with bone marrow of type 2 diabetes. DESIGN, TIME AND SETTING: The randomized control experiment was performed at the Center Laboratory of the Third Xiangya Hospital, Central South University between August and December 2007. MATERIALS: Thirty male Sprague Dawley rats were divided into the control (n=10) and experimental (n=20) groups, fed with normal diet and high fat and sucrosum diet, respectively. METHODS: After 8 weeks of feeding, blood of rats were collected by tail cutting method. And rats were intraperitoneally injected with 30 mg/kg streptozotocin or same volume citrate buffer. After 2 weeks, the same way was used to collected blood. Models of distraction osteogenesis in the left tibias of the rats were established. All animals received osteotomy and placement of the external fixators on the lift tibiae for 14 days. MAIN OUTCOME MEASURES: Callus formation in distraction gap was compared by X-ray. The formation of primary matrix front and microcolumn formation was observed by histological examination, then calculated the adipocytes accounting. The expression of PPAR γ and CBF α 1 mRNA in femurs marrow was measured by RT-PCR. RESULTS: After feeding with high fat and sucrosum diet, the level of total cholesterol, triglycedde and fasting blood glucose was obviously elevated compared with control group (P 〈 0.05 0.01 ). After 2 weeks of administration, the concentration of triglyceride, total cholesterol, and fasting blood glucose was elevated than that of the control group (P 〈 0.05 0.01
