摘要
目的探讨低氧诱导因子-1α(HIF-1α)对成骨细胞增殖、分化和矿化功能的影响。方法分别在21%和2%O2浓度下培养成骨细胞,21%O2培养24 h和2%O2培养6、12、24 h后分别检测成骨细胞表达HIF-1α、血管内皮生长因子、核结合因子a1、骨钙素的水平,检测成骨细胞增殖率、分泌碱性磷酸酶情况以及钙结节形成情况。结果与21%O2培养相比,2%O2浓度下,成骨细胞在mRNA和蛋白水平HIF-1α表达均上升,同时成骨细胞表达血管内皮生长因子、核结合因子a1、骨钙素的水平逐渐升高;成骨细胞增殖率增加,分泌碱性磷酸酶的能力以及钙结节形成能力也逐渐增强。结论在2%O2浓度下24 h内,HIF-1α能促进成骨细胞增殖、分化和矿化,从而促进成骨细胞的成骨功能。
Objective To investigate the regulatory effect of hypoxia-inducible factor -1α (HIF-1α) on the proliferation, differentiation and mineralization of osteoblast. Methods Osteoblasts were cultured under 21% O2 for 24 h and 2% O2 for 6, 12 and 24 h. RT-PCR was used to detect the mRNA expression of HIF-1α, vascular endothelial growth factor (VEGF), core binding factor α1 (RunX2) and osteocalcin(OC). Western-blot was employed to detect the protein expression of HIF-1α, VEGF and RunX2. Alkaline phosphatase (ALP) bioactivity, bone nodule formation and proliferation of osteoblasts were also detected. Results With the augmentation of the expression of HIF-1α and VEGF on mRNA and protein level under 2% O2, the expression of RunX2 and OC, the ALP bioactivity, the amount of bone nodule formation and the ability of osteoblast proliferation were increased compared with 21% O2. Conclusion HIF-1α can promote the proliferation, differentiation, mineralization of osteoblasts and enhance the ability of osteogenesis under 2% O2 within 24 h.
