摘要 目的 探讨腺病毒介导的核心结合因子a1(corebindingfactora1,Cbfa1)基因转移对NIH3T3细胞的调节作用。方法 含有Cbfa1基因的重组腺病毒转染NIH3T3细胞,RT PCR检测成骨标志基因骨钙素(osteocalcin ,OCN)、Ⅰ型胶原(typeⅠcollagen)、骨唾液蛋白(bonesialoprotein ,Bsp)、碱性磷酸酶(alkalinephosphatase ,AKP)、Cbfa1的表达。Westernblot检测Cbfa1的表达,免疫细胞化学染色检查OCN的表达。结果 NIH3T3细胞在Cbfa1基因转移后RT PCR检测到OCN、Bsp、AKP、typeⅠcollagen基因的表达,Westernblot检测到Cbfa1蛋白的表达,免疫细胞化学染色检测到了OCN蛋白的表达。结论 腺病毒介导Cbfa1基因转移NIH3T3细胞后出现成骨表型。 Objective To study the modulation effect of adenovirus-transcripted transfer of core binding factor a1 (Cbfa1) gene on NIH3T3 cells. Methods After the adenovirus-transcripted transfer of Cbfa1 gene in NIH3T3 cells,the expression of osteocalcin (OCN), type Ⅰ collagen, bone sialoprotein (Bsp) and alkaline phosphatase(AKP) was determined with RT-PCR. The expression of Cbfa1 gene was screened with Western blotting. The expression of OCN was examined with immunocytochemical staining. Results After the transfer of Cbfa1 gene, the expression of type Ⅰ collagen, OCN, Bsp and AKP was detected with RT-PCR. Expression of Cbfa1 was identified with Western blotting. The expression of OCN was confirmed with immunocytochemical staining. Conclusion The transfer of Cbfa1 gene results in the appearance of the osteogenetic phenotype of NIH3T3 cells.
