重组人胰岛素样生长因子2对豚鼠眼球发育的调控作用及其与形觉剥夺的关系 被引量:2

摘要 目的对豚鼠非遮盖眼和形觉剥夺性近视(form-deprivation myopia,FDM)眼玻璃体腔注射重组人胰岛素样生长因子2(recombinant human insulin-like growth factor,rhIGF-2),观察rhIGF-2对两组豚鼠眼屈光度、眼轴长度及视网膜-脉络膜内源性IGF-2表达水平的影响,探讨rhIGF-2对豚鼠眼球发育的调控作用以及该调控作用与形觉剥夺的关系。方法选取3周龄的三色豚鼠80只,随机均分为2组。A组为非遮盖组,分为A1~A5组,每组8只:A1组为空白对照组;A2组为阴性对照组,单眼玻璃体腔注射20μl牛血清白蛋白;A3~A5组为rhIGF-2注射组,注射量分别为1ng、10ng、100ng(20μl)。B组为单眼遮盖组,分为B1~B5组,各组遮盖眼处理同A1~A5组。实验第14天测量遮盖眼屈光度和眼轴长度,检测视网膜-脉络膜IGF-2的表达水平。结果非遮盖组中,rhIGF-2注射组(A3、A4、A5组)的屈光度、眼轴长度与对照组(A1、A2组)之间差异无统计学意义(P>0.05);但A4、A5组的IGF-2水平明显低于对照组(P=0.000),A3组与对照组差异无统计学意义(P>0.05);A3、A4、A5组内源性IGF-2表达水平与rhIGF-2注射量呈负相关(r=-0.940,P=0.000)。单眼遮盖组中,B4、B5组与对照组(B1、B2组)相比,近视屈光度增加,眼轴延长,内源性IGF-2表达水平下降(P=0.000),而B3组与对照组之间差异无统计学意义(P>0.05);B3、B4、B5组的屈光度及内源性IGF-2表达水平与rhIGF-2注射量呈负相关(r=-0.832,-0.858,P=0.000),眼轴长度与rhIGF-2注射量呈正相关(r=0.863,P=0.000)。结论rhIGF-2玻璃体腔注射对豚鼠非遮盖眼的屈光度和眼轴长度无影响,但可使形觉剥夺眼近视屈光度增加,眼轴延长,其效应呈浓度依赖性。rhIGF-2可下调豚鼠眼视网膜-脉络膜内源性IGF-2的表达,效应呈浓度依赖性。提示rhIGF-2在形觉剥夺存在的条件下可促进形觉剥夺性近视的发展。 [Abstract] Objective To clarify the effects of recombinant human insulin-like growth factor (rhIGF-2) on the development of guinea pig eyes and the relationship between its function and form- deprivation (FD). Methods Eighty 3-week-old guinea pigs were included in the study. They were randomly divided into 2 groups: group A (n=40) was the non-FD group. Group A was further divided into 5 subgroups: group A1 (n=8) was the control group, group A2 (n=8) was the negative control group that received an intravitreous injection of 20 μl bovine serum albumin (BSA); groups A3~A5 (n=24) received intravitreous injections of 1 ng, 10 ng and 100 ng(20 μl) rhIGF-2. Group B (n=40) was the FDM group. Group B was further divided into subgroups B1~B5 using the same criteria as group A. The refraction and axial length of all guinea pigs were measured at day 7 and the expression of IGF-2 was assayed by Western-blot. Results In the non-FD group, the rhIGF-2 injection did not have significant effects on either refraction or axial length (F=0.217, 0.562; P=0.928, 0.692), while there was a significant difference in the level of IGF-2 expression in groups A3~A5 (P=0.000) and there was a negative correlation between the level of IGF-2 expression and the concentration gradient of rhIGF-2 (r=-0.940, P=0.000). In the FDM group, the degree of myopia and axial length increased in group B4 and group B5 compared to that of groups B1 and B2. The degree of myopia and the level of IGF-2 mRNA of groups B3~B5 had a negative correlation with the concentration gradient of rhIGF-2 (r=-0.832, -0.858, P=0.000), while axial length had a positive correlation (r=0.863, P=0.000). Conclusion The intravitreous injection of rhIGF-2 has no effect on the degree of myopia and axial length of the non-FD guinea pig, while it can induce significant increases in the degree of myopia and axial length in the FDM guinea pig and the effects are concentration dependent.
