关节软骨钙化层界线结构特异性染色方法建立 被引量:1

摘要 目的建立关节软骨钙化层界线结构特异性染色方法。方法从组织库获取成人股骨髁标本10例,修剪、固定后常规方法制备石蜡切片,联合应用HE、天狼猩红、番红O/固绿和冯库萨等染色方法,对关节软骨钙化层界线结构进行观察。结果HE染色仅能依据各层组织的结构、形态区分3层结构,两界线显示不清;天狼猩红染色和番红O/固绿染色可以清晰区分粘合线结构,但潮线界面显示不清;冯库萨染色可以清晰显示钙化软骨层两界线结构,但软骨下骨的形态结构显示不清。结论通过对不同方法染色结果进行比较,番红O/固绿染色和冯库萨染色联合应用可以清晰显示钙化层界线结构。 Objective To found the specific staining method for the limit structure of articular calcified cartilage zone. Methods The normal human condyles of femur (n=10) were donated by the residents of Chong Qing city from tissue bank,and the paraffin sections were prepared after fixation and decalcification. Observed the limit structure of articular calcified cartilage zone under the microscope after stained with HE,Picrosirius,Safranin O/fast green and von kossa. Results Three-layered structures of the articular bone-cartilage were distinguished according the structure and shape with HE stain,but the two limit structures could not be observed clearly. The cement line was distinct with the Picrosirius stain or the Safranin O/fast green stain,but the tidemark could not be observed clearly. The tidemark was distinct with the von kossa stain, but cement line could not be observed clearly. Conclusion The limit structure of articular calcified cartilage zone can be observed clearly with the Safranin O/fast green stain and von kossa stain.
