RP-HPLC双波长法测定白芍中芍药苷和五没食子酰葡萄糖含量 被引量:15

摘要 目的建立测定白芍药材中芍药苷和五没食子酰葡萄糖含量的双波长RP—HPLC分析方法。方法采用HypersilODS(4.6×250mm,5μm)色谱柱,以乙腈和0.05%磷酸为流动相,梯度洗脱,流速1.0mL/min,检测波长为230nm和275nm,柱温25℃。结果芍药苷在0.208~2.08μg呈良好的线性关系,r=0.9999,平均回收率96.3%;五没食子酰葡萄糖在0.04~0.4μg呈良好的线性关系,r=0.9998,平均回收率98.9%。结论该方法简便、快速、准确、灵敏度高,可用于白芍的质量控制。 Objective To establish a dual wavelength method of RP-HPLC for the quantitative determination of paeoniflofin and pentagalloylglucose in Paeonia lactiflora Pall. Methods The dual wavelength method of RP-HPLC was performed on a column of Hypersil ODS (4. 6 mm × 250 mm, 5 μm), using acetonitrilephosphoric acid solution (0.05% ,V/V) as the mobile phase in gradient elution. The flow rate was 10 mL/ min, detection wavelength was 230 nm and 275 nm respectively, and the temperature was 25℃. Results Paeoniflorin showed a good linear relationship at range of 0. 208 μg - 2.08 μg ( r = 0. 999 9 ). The average recovery was 96. 3 %. Pentagalloylglucose showed a good linear relationship at range of 0. 04 μg- 0. 4μg (r =0. 999 8). The average recovery was 98.9 %. Conclusion The method is simple, convenient, accurate and sensitive, which can be used for the quality control of Paeonia lactiflora Pall.
