摘要
目的探讨单侧帕金森病(PD)模型大鼠黑质中内源性孤儿核受体(Nurr1)和酪氨酸羟化酶(TH)mRNA表达的变化及其相关性。方法用6-羟基多巴胺(6-OHDA)注射至成年大鼠右侧前脑内侧束(MFB)制备单侧帕金森病模型,于术后1、7、14、28 d分别采用半定量RT-PCR和原位杂交方法检测双侧黑质中Nurr1和TH mRNA表达的变化。结果Nurr1 mRNA表达在术后1 d损伤侧黑质中即明显下降,术后7 d开始逐渐增加,14 d接近正常水平,28 d明显升高。除14 d组与正常对照组相比,差异无统计学意义(P>0.05)外,其余各组间的差异均有统计学意义(均P<0.05)。而TH mRNA表达在术后1 d损伤侧黑质中无明显变化,7 d后开始下降,14 d达最低,28 d开始上升。除1d组与正常对照组相比,差异无统计学意义(P>0.05)外,其余各组间的差异均有统计学意义(均P<0.05)。结论Nurr1mRNA在多巴胺能表型神经元的损伤和再生修复中发挥重要的作用。
Objective To study the change and relativity of Nurr1 and TH mRNA expression in the substantia nigra of unilateral Parkinson’s disease(PD) model rats.Methods 6-OHDA was injected into the right MFB of adult rats to construct unilateral PD model,semi-quantitative RT-PCR and in situ hy-bridization were used to detect the change of Nurr1 and TH mRNA expression in bilateral substantia nigra on the 1st,7th,14th and 28th day respectively after operation.Results The Nurr1 mRNA ex-pression apparently decreased in the leisoned substantia nigra on the 1st day after operation,started to increase gradually on the 7th day,approched the normal level on the 14th day,and markedly increased on the 28th day.However,the TH mRNA expression was of little change in the leisoned substantia ni-gra on the 1st day after operation,started to decrease on the 7th day,reached the bottom on the 14th day,and started to increase on the 28th day.Conclusion The Nurr1 mRNA may play an important role in the injury and restorative process of dopaminergic neurons.
