摘要
目的观察单核细胞趋化蛋白-1(MCP-1)在多柔比星肾病大鼠肾组织及尿液中的表达,探讨MCP-1在微小病变肾病综合征(MCNS)中的作用机制。方法雄性Wistar大鼠83只。随机分为正常对照组(n=40)和肾病组(n=43)。大鼠尾静脉单次注射多柔比星5mg/kg制备模型。在实验第7、14、28、42天检测各组大鼠24h尿蛋白定量,同时采用ELISA检测各组大鼠尿、肾组织MCP-1水平,采用SPSS11.0软件对各组间差异进行t检验和两变量间的相关性分析。结果1.肾病组大鼠尿蛋白排泄量逐渐增加,第7、14、28、42天与正常对照组比较,均有显著性差异(Pa<0.01)。2.不同时期肾病组大鼠肾组织及尿液MCP-1均明显高于同期正常对照组(Pa<0.01)。3.不同时期肾脏及尿液中MCP-1的变化分别与24h尿蛋白定量呈正相关(r=0.583,0.651Pa<0.01)。尿液MCP-1水平与肾脏MCP-1水平也呈正相关(r=0.810P<0.01)。结论MCP-1参与了多柔比星肾病大鼠蛋白尿的发生。尿液中MCP-1水平可反映肾组织MCP-1水平,可作为判断MCNS病情的检测指标之一。
ObjectiveTo observe the expression of monocyte chemoattractant protein-1（MCP-1）in renal tissue and urine of Doxorubicin-induced nephropathy rats,and to discuss its possible mechanism in minimal change nephrotic syndrome（MCNS）.MethodsEighty-three male Wistar rats were enrolled in this study.They were randomly divided into normal control group（n=40） and nephropathic group（n=43）.The nephropathic group was established by a single intraperitoneal injection of Doxorubicin 5 mg/kg.The 24 h urine protein of all rats was measured on the 7th,14th,28th and 42th day.The MCP-1 level in renal tissue and urine were determined with ELISA.SPSS 11.0 software was used to obtain t-test of various groups and correlation analysis of 2 variables.Results1.The 24 h urine protein of nephropathy rats increased gradually on the 7th,14th and 28th day,and it was greatly higher than that of normal control group（Pa〈0.01）.2.The MCP-1 level in renal tissue and urine of nephrotic group each time point was significantly increased than that of the normal control group（Pa〈0.01）.3.The MCP-1 in urine and 24 h urine protein concentrations were positively correlated（r=0.651P〈0.01）.The MCP-1 level in renal tissue and 24 h urine protein concentrations were positively correlated（r=0.583P〈0.01）.In kidney and urinary concentrations of MCP-1 was positively correlated（r=0.810P〈0.01）.ConclusionsMCP-1 involved in the incidence of proteinuria in rats with Doxorubicin-induced nephropathy.MCP-1 level may reflect the MCP-1 level of renal tissue,and as an index to judge MCNS condition.
