摘要 目的:确定银屑病皮损组织与非皮损组织中微小RNA(miRNA)表达的差异。方法:从寻常型银屑病皮损组织和非皮损组织取材,Trizol法抽提细胞总RNA,Ambion miRNA Isolation Kit进一步分离miRNA;利用基因芯片技术,将细胞miRNA与哺乳动物miRNA芯片杂交,采用Lux-Scan 3.0图像软件和SAM version 2.1进行数据分析。结果:银屑病皮损组织与非皮损组织表达的miRNA中有84个存在着明显差异(fold change>2.0),其中46个低表达和38个高表达,最高差异达36倍,部分miRNAs与细胞增殖关系密切。结论:银屑病皮损组织较非皮损组织中miRNA表达数量更丰富,细胞代谢和增殖更活跃;部分miRNAs可能参与银屑病表皮细胞增殖的分子机制。 Objective: To determine the differential expression of miRNA (microRNA) between the lesional tissue and non- lesional tissue of psoriasis vulgaris. Methods: The total RNAs in the lesional tissue and non- lesional tissue of psoriasis vulgaris patients were extracted by Trizol and miRNAs were further isolated by Panbion' s miRNA Iso- lation Kit. Hybridization of cell miRNAs to mammalian miRNA chip was made. Data analysis was proceeded by software of LuxScan3.0 and SAM version2.1. Results: A total of 84 miRNAs were found, including 46 low - expression and 38 over- expression miRNAs. The difference between the highest and lowest expressions was 36 folds. Data analysis revealed that some miRNAs were associated with epidermal cell proliferation. Conclusion: The miRNAs expression is more abundant in the lesional tissue than in the non - lesional tissue of psoriasis, reflecting the active metabolism and rapid proliferation in psoriasis. Some miRNAs may be involved in the pathogensis of psoriasis.
