摘要
目的探讨miRNA-210在寻常型银屑病皮损中的表达情况及影响。方法收集临床银屑病患者的皮损组织和健康皮肤组织,经过速冻后提取miRNA。采用Stemp-loop技术反转录miRNA,采用Real time PCR技术检测miRNA的表达,采用绝对定量方法计算miRNA-210的拷贝浓度。免疫组化法对寻常型银屑病皮损组织及正常皮肤组织中RUNX3蛋白的表达结果进行对比分析,并分别记录。结果 Real time PCR显示miRNA-210在寻常型银屑病患者组皮损中表达量较健康组皮肤显著降低(P<0.01);免疫组化结果显示RUNX3蛋白表达阳性率,寻常型银屑病患者皮损中显著高于健康组皮肤(P<0.05)。结论寻常型银屑病患者皮损中miRNA-210显示为低表达,其可能通过调节靶基RUNX3,参与银屑病发病环节。
Objective To explore the expression of miRNA-210 in the lesions of psoriasis and the influence of the disease.Methods The skin tissue of clinical patients with psoriasis and the healthy were collected.The miRNA was extracted by us after quick-frozen.The Stemp-loop technology was used to reverse transcription of miRNA and Real time PCR tevhnique was applied to identity the expression of miRNA.The standard curve should be made and the miRNA-210 copies of concentration be calculated by absolute quantitative method.The t-test was used to compare the difference between psoriatic skin lesions tissues and the normal skin tissues,and A p-value less than 0.05 was considered statistically significant.The expression of RUNX3 in the two tissue groups were determined with immunohistochemical method.Results Real time PCR showed that the expression of miRNA in patients with psoriasis vulgaris groups decreased significantly compared with the normal groups(P〈0.05).The results of immunohistochemistry demonstrated that the expression of RUNX3 protein in skin lesions of patients with psoriasis vulgaris were significantly higher than that normal skin(P〈0.05).Conclusion Lower expression of miRNA-210 was detected in the lesions of patients with psoriasis vulgaris.The miRNA-210 may effect on its target genes RUNX3 and then play a role in the onset of psoriasis.
