摘要
目的观察不同剂量水杨酸钠(sodiumsalicylate,NaSA)对抗顺铂(cisplatin,CDDP)所致听损伤的作用。方法60只健康成年豚鼠随机分为4组,每组15只。分别为:Ⅰ组:CDDP+NaSA(50 mg/kg)、Ⅱ组:CDDP+NaSA(100 mg/kg)、Ⅲ组:CDDP+NaSA(150 mg/kg)、Ⅳ组(对照组):CDDP+生理盐水(NS)组。观察用药前后各组听性脑干反应(auditory brainstemresponse,ABR)阈值及畸变产物耳声发射(DPOAE)幅值的变化。结果用药后Ⅰ、Ⅱ、Ⅲ组与Ⅳ组相比,Ⅳ组ABR阈值升高较其他3组明显,差异有显著统计学意义(P<0.05)。Ⅰ、Ⅲ组分别与Ⅱ组相比,ABR阈值差异无显著统计学意义(P>0.05),而Ⅰ组ABR阈值明显高于Ⅲ组(P<0.05)。Ⅰ、Ⅱ、Ⅲ组在1、2、3、4、6、8 kHz各频率DPOAE幅值明显大于Ⅳ组(1、3 kHz,P<0.05;2、4、6、8 kHz,P<0.01)。结论水杨酸钠对顺铂所致的听损伤具有明显拮抗作用。
Objective To investigate the antagonist effects of sodium salicylate (NASA) on the cisplatin (CDDP) induced hearing impairment in guinea pigs. Methods 60 guinea pigs were randomly divided into four groups: ① CDDP +NASA(50 mg/kg) group, ②CDDP +NASA(100 mg/kg) group ,③CDDP +NASA(150 mg/kg) group and ④CDDP +NS(normal saline) control group. Auditory brainstem response (ABR) and distortion product otoacoustic emission (DPOAE) were used to evaluate their effects on hearing threshold and DPOAE amplitudes. Results The ABR responses of groups Ⅰ , Ⅱ and Ⅲ were significantly lower than in the control group (P〈0.01) while they were insignificantly different in group Ⅱ from the other two groups (P〈0.05). The ABR responses for group Ⅰ were significantly higher than group Ⅲ (P〈0.05). The amplitudes of DPOAE at 1, 2, 3, 4, 6 and 8 kHz in group Ⅰ , Ⅱ and Ⅲ were greater than those in the control group (1,3 kHz, P〈0.05; 2, 4, 6, 8 kHz,P〈0.01). Conclusion Sodium salicylate can effectively reduce ototoxicity of cisplatin.
