摘要
目的探讨阿司匹林对卵巢癌细胞凋亡的影响及对凋亡相关基因的作用。方法培养卵巢癌细胞株SK-OV-3,用不同浓度阿司匹林进行干预,用Giemsa染色观察细胞形态学改变,流式细胞仪方法定量检测细胞的凋亡及细胞周期的变化,用RT-PCR法检测凋亡相关基因Bcl-2/bax表达。结果阿司匹林对SK-OV-3增殖的抑制率随作用时间的延长和药物浓度的增加而增加;阿司匹林作用后SK-OV-3细胞出现了典型的凋亡形态学变化,可见凋亡小体;不同浓度的阿司匹林处理细胞48 h后,流式细胞术(FCM)显示细胞的凋亡呈剂量依赖性,1、5、10 mmol/L阿司匹林诱导SK-OV-3细胞的凋亡率分别为(11.1±0.5)%、(17.9±0.4)%、(30.8±0.4)%均高于对照组(P<0.01);同时,随着阿司匹林浓度的增加,G0/G1期细胞比例下降,G2/M期细胞比例升高,细胞被阻断在G2/M期,细胞周期发生明显改变;阿司匹林作用后的卵巢癌细胞的凋亡相关基因Bcl-2表达降低,bax基因表达升高。结论阿司匹林能抑制卵巢癌细胞增殖,诱导其凋亡,且阿司匹林诱导SK-OV-3细胞凋亡是通过下调Bcl-2表达和上调bax表达,而达到抗肿瘤的作用。
Objective To investigate the effect of aspirin on apoptosis of ovarian carcinoma cells and expression of Bcl-2/bax mRNA. Methods Morphologie changes of apoptotie eells were observed by optical microscope after Giemsa staining. Cell apoptosis and cell cycle was detected by cell flow eytometry (FCM). The expression of Bcl-2/bax mRNA was detected by RT-PCR. Results Morphology changes of apoptosis and apoptotie body were found by optical microscope. The apoptosis percentage (AP) induced by aspirin was detected by FCM, which showed that aspirin at dosage of 1,5,10 mmol/L for 48 h eould induce cell apoptosis in a dose-dependent way ranging from (11.1 ± 0.5)%, ( 17.9±0.4) %, (30.8±0.4) % respectively ( P 〈 0.01 ). There were visible apoptotie peaks of hypodiploid on the DNA bargraph. Compared with control group, after treated with aspirin, SK-OV-3 cells of G0/G1 stage decreased and cells of G2/M stage increased. Aspirin could down-regulate the expression of Bcl-2 mRNA, up-regulate the expression of bax mRNA. Conclusion Aspirin can inhibit the proliferation of ovarian carcinoma cells and induce its apoptosis, and whieh can induce the apoptosis of SK-OV-3 cells by down-regulating the expression of Bcl-2 mRNA, and up-regulating the expression of bax mRNA, so that aspirin has the effect of antineoplasna.
