摘要
应用体外iTRAQ标记结合LC-MALDI-TOF-TOF方法对雌二醇刺激、雌二醇与它莫西芬共同处理,将它莫西芬处理与正常培养的MCF-7乳腺癌细胞内蛋白质表达的差异进行了比较分析。结果表明:各实验组与对照组之间存在数种上调或下调1倍以上的差异表达蛋白质,雌二醇可显著改变74种蛋白质的含量,雌二醇与它莫西芬共同处理引起26种蛋白质水平的显著变化,它莫西芬引起18种蛋白质表达的明显改变。各组之间既有变化趋势相同的蛋白质,也有变化趋势相反的蛋白质。
The differential expression of intracellular proteins from 17/3-oestradiol(E2) stimulated, E2 plus tamoxifen(TAM) treated or TAM treated MCF-7 breast cancer cells was analyzed and compared with that from regularly cultured MCF-7 cells by using iTRAQ labeling coupled with LC - MALDI- TOF-TOF strategy. Numerous proteins were discovered to be significantly upregulated or downregulat- ed by more than two folds. E2, E2 + TAM or TAM altered the expression of 74, 26 and 18 proteins significantly. The comparison of the change trends of protein between experimental groups and control groups showed that some proteins change to the same direction while others change to reverse direction. This study provides some useful information for elucidating the molecular mechanisms for the effects of E2 and TAM on breast cancer cells on protein level.
