摘要 iTRAQ是2004年开发的一种新的同位素标记试剂,结合LC-MS/MS技术已被广泛地应用于细菌、酵母、人体组织、细胞和体液等多种样本的蛋白质鉴定和定量。作为一种新的定量蛋白质组学研究方法,该技术可同时对4种样本进行定量分析并展现了高通量、重复性好、敏感性高等特点。iTRAQ技术为我们研究前列腺癌的发生、进展机制,发现新的生物学标记物提供了有力的技术平台。 Amine-specific isobaric tagging (iTRAQ) reagents,as a new class of isobaric reagent,were developed in 2004,which,combined with liquid chromatography and tandem mass spectrometry (LC-MS/MS),have been applied to the identification and quantification of proteins in a wide range of biological samples,including bacteria,yeasts,human tissues,cells,and fluids. As a new method of quantitive proteomics,the technique of iTRAQ allows for the quantitive analysis of four samples simultaneously and displays its advantages of high-flux,food reproducibility,and high sensitivity; it also provides a potential technological platform for studying the mechanisms of the development and progression of prostate cancer.
