摘要
目的：观察瑶药猛老虎提取物对大鼠肝星状细胞（HSC）Ⅰ、Ⅲ型胶原、基质金属蛋白酶（MMP-1）和转化生长因子-β1（TGF-β1）表达的影响。方法：制备猛老虎乙酸乙酯部位分离物药物血清，分离培养大鼠肝HSC，温育HSC，EIJSA法检测上清液中Ⅰ、Ⅲ型胶原含量；MTT检测HSC的生长情况；逆转录多聚酶链反应（RT—PCR）法观察MMP-1的表达；免疫组化法观察TGF-β1的表达。结果：猛老虎乙酸乙酯部位各分离物和秋水仙碱药物血清对体外培养的HSC细胞增殖无抑制作用。与空白对照血清组比较，无显著性差异（P〉0．05）。分离物E中和高剂量组的药物血清均能明显促进体外培养HSC细胞MMP-1基因表达水平、抑制Ⅰ、Ⅲ型胶原的分泌和TGF-β1表达并呈现剂量依赖关系，与空白对照血清组比较，均具有显著性的统计学意义（P〈0．05或P〈0．01），效果与秋水仙碱相当。结论：瑶药猛老虎抗肝纤维化作用不是直接抑制HSC的增殖，而是与其促进HSC细胞MMP-1基因表达水平、抑制Ⅰ、Ⅲ型胶原的分泌和TGF-β1的表达有关。
Objective： To investigate the effect of extract of Yao Nationality herb named ＂MengLaoHu＂on expression of collagen - Ⅰ, collagen - Ⅲ,matrix metalloproteinases - 1 （MMP - 1 ） and transforming growth factor - β1 （TGF -β1 ） in rat＇s hepatic satellite cells（HSC）.Methods：Preparated the MengLaoHu of ethyl acetate extract and rats＇ blood serum collected to prepare the medicated serum, which was incubated with the subcultured HSC. HSC were isolated and cultured from the liver of rats and were incubated. Collagen - Ⅰ and collagen -Ⅲ content in the medium were measured with EIJSA. Growth conditions of HSC were measured by means of Methylthio tetrazole（MTT）method. The expressions of MMP- 1 were determined by reverse transcription- polymerase chain reaction（RT- PCR）. Immuno- histochemistry was used to detect the expression of TGF-β1 .Results： The genomie variation of the ＂MengLaoHu＂ of ethyl acetate extract and drug sera of colchicines had no inhibit the proliferation of HSC. Compared to blank control group. There was no significant difference（ P 〉 0.05）. E0.26 isolates and the group of 0.78g/kg medicated sera can obviously promote the levels MMP- 1 genes expression, can down- regulate the expression of Ⅰ , Ⅲ - type collagen and TGF-β1, and the effect was in a dose - dependent manner. The difference being significant as compared with the normal control group （ P 〈 0.05 or P 〈 0.01）, identical as colchicines. Conclusions： ＂MengLaoHu＂ is not directly inhibited the proliferation of HSC,but related to promote the levels MMP- 1 genes expression, down - regulate the expression of Ⅰ , Ⅲ- type collagen and TGF- β1.
