糖尿病太鼠痛性周围神经病变时脊髓小胶质细胞的活化 被引量:3

Change in microglia activation in spinal cord in a rat model of diabetic neuropathic pain

摘要 目的评价糖尿病大鼠痛性周围神经病变时脊髓小胶质细胞的活化。方法SD大鼠25只,2月龄,雌雄不限,腹腔注射1%链脲佐菌素60mg/kg以制备大鼠糖尿病痛性周.围神经病。变模型,取造模成功的10只SD大鼠作为糖尿病痛性周围神经病变组(DM组),另取10只同月龄SD大鼠作为对照组(NC组)。分别于注射前(T1)、注射后2、7、14、21、28d(T2~6)时称量体重,取尾静脉血0.1ml测定血糖水平,于T1、T3-6时测定大鼠右后爪机械缩足反应阈值;注射28d时麻醉大鼠,取L4,5脊髓制备切片,采用免疫组化法检测脊髓小胶质细胞补体受体3(CR3)的表达。结果与NC组比较,DM组T2~6时血糖升高、体重下降,T4-6时机械缩足反应阈值降低,T6时小胶质细胞CR3表达上调(P〈0.05或0.01);与T1时比较,DM组L2-6时血糖升高、体重下降,T6时机械缩足反应阈值降低(P〈0.01)。结论脊髓小胶质细胞的活化与大鼠糖尿病痛性周围神经病变的发病有关。 Objective To investigate the change in microglia activation in the spinal cord in a rat,model of diabetic neuropathie pain. Methods Twenty 2-month old SD rats of both sexes weighing 180-200g were divided into 2 groups( n = 10 each): group Ⅰ normal control (NC) and group Ⅱ diabetes mellitus (DM). Diabetes mellitus was induced by intraperitoneal (IP) streptozotocin (STZ) 60 mg/kg and confirmed by blood glucos level〉 16.7 mmol/L. Body weight and blood glucose were measured before (T1) and at 2, 7, 14, 21, 28 day after STZ IP injection (T2-6). Paw withdrawal threshold to van Frey filament stimulation was measured at T1 and T3-6 . The animals were killed and lumbar segments of the spinal cord were removed for determination of expression of complement receptor 3 (CR3) in mieroglia in the spinal cord by immuno-histoehemieal assay. Results The blood glucose level was significantly increased While body weight was significantly decreased after IP STZ injection as compared with the baseline before STZ injection in group DM. The blood glucose level was significantly higher and body weight significantly lower after STZ injection in group DM than in group NC. The Paw withdrawal threshold was significantly decreased after STZ at T4-6 as compared with the baseline at T1 in group DM and was significantly lower than in group NC. Immuno-histoehemistry demonstrated that the expression of CR3 in mieroglia in the dorsal horn was significantly up-regulated after STZ injection in group DM. Conclusion Mieroglia activation is involved in the pathogenesis of diabetie neuropathie pain.
