摘要
目的:观察糖尿病痛性周围神经病变大鼠脊髓星形胶质细胞的变化。方法:采用腹腔注射链尿佐菌素(STZ)诱导糖尿病痛性周围神经病变大鼠模型,在造模成功的痛敏糖尿病大鼠中随机取10只作为糖尿病组(Ⅱ组),另取10只同月龄大鼠为空白对照组(I组)。于注射STZ前、注射STZ后2d、7d、14d、21d、28d测量大鼠体重并取尾静脉血测定血糖;注射STZ前、注射STZ后7d、14d、21d、28d用von Frey丝测定50%缩足反应阈值(PWT);注射STZ28d后处死大鼠,采用免疫组化的方法检测大鼠脊髓的神经胶原纤维酸性蛋白(GFAP)。结果:与I组相比,注射STZ后Ⅱ组血糖显著增高(P<0.01),体重逐渐下降;注射STZ后21-28dⅡ组PWT明显低于I组(P<0.05);注射STZ后第28d检测大鼠脊髓的GFAP的变化:与I组相比,Ⅱ组明显增多(P<0.05)。结论:糖尿病痛性周围神经病变大鼠脊髓星形胶质细胞增殖活化,可能参与了糖尿病周围神经痛的产生和维持。
Objective: To evaluate the astroglial response in spinal cord in the streptozotocin (STZ)-induced diabetic rat model with painful diabetic neuropathy. Methods: Painful diabetic neuropathy was induced by intraperitoneal injection of STZ in 10 Sprague-Dawley rats ( group Ⅱ), 10 age-matched healthy rats were served as control group (group Ⅰ). The blood samples were collected from vena caudalis to assess the blood glucose level before STZ injection and 2d,7d, 14d,21d,28d after STZ injection. Body weights were measured at the same time points. Responses to the mechanical stimulus were measured with von Frey filament, and 50% paw withdraw threshold (PWT)was recorded before STZ injection and 7 d, 14 d,21d,28d after STZ injection. At 28d after STZ injection, the animals were sacrificed and lumbar segments of spinal cord were collected for analysis of glial fibrillary acidic protein (GFAP) in the astrocytes in spinal cord by immuno-histochemical assay. Results: The level of blood glucose increased dramatically after STZ injection in group Ⅱ while the body weight decreased. The PWT in group Ⅱdecreased significantly comparing to group Ⅰ at 21 -28 d after STZ injection. Immuno-histochemical assay demonstrated that GFAP positive astrocytes in the dorsal horn significantly increased at 4W after STZ injection. Conclusion: Astrocytes activation may be involved in the pathogenesis of painful diabetic neuropathy.
