摘要
目的研究大肠癌组织中FⅦ的表达情况,探讨FⅦ表达量和大肠癌、肿瘤肝转移的相关性。方法以北京大学第一医院新近(2006年1月至2007年6月间)手术切除的大肠癌肿瘤标本和同一病例的大肠正常黏膜取材,共30例。通过免疫组织化学方法检测FⅦ在大肠癌组织当中的合成情况。通过Western—Blot方法检测FVU在其中18例不同Dukes分期的大肠癌组织当中合成量的差异。结果免疫组织化学法发现在大肠癌组织当中有FⅦ高表达(17/30),Western—Blot蛋白质印迹法证明在不同Dukes分期的大肠癌肿瘤组织中的FⅦ表达存在差异,从B期到D期,表达量逐渐提高,尤其是D期与其他各期存在明显差异。结论大肠癌组织中存在异源性的活性凝血因子Ⅶ蛋白的合成;大肠癌组织中的FVR表达量和肿瘤的Dukes分期密切相关,可能具有促进大肠癌肿瘤的侵袭和转移能力。
Objective To study the synthesis of clotting factor Ⅶ in colorectal cancer, and the correlation between its expression volume and liver metastasis. Methods Colorectal cancer tissue and normal tissues of colon were taken from recent 30 operated patients. The protein synthesis of FⅦ in cancer tissue was verified by immunohistochemistry and its expression volume variance was compared by Western blotting. Results Colorectal cancer tissue can synthesize FⅦa protein, however, it is negative in normal colorectal tissues. Western blotting reveals the variance of FⅦa protein expression in colorectal cancer in different Dukes stages. The expression volume increases with increasing stage B to D, especially in stage D. The FⅦ volume in stage D is significantly higher than those in the other 2 stages. Conclusions Colorectal cancer tissue can synthesize active FⅦa. The expression volume of FⅦ correlates with the Dukes stages, and the FⅦ activity may enhance the invasive and metastatic capability of the colorectal tumor.
