摘要 目的从龙眼肉中提取分离粗多糖LYRP,测定纯化后的组分分子质量及其单糖成分。方法采用超声浸提法、酶水解法去蛋白质、DEAE-纤维素和SephacrylS-300柱层析法得到多糖组分LYRP2-1。高效凝胶渗透色谱测定分子质量,完全酸水解得到单糖成分,紫外光谱和傅立叶转换红外光谱法对其定性分析。结果均一LYRP2-1具有多糖特征性的紫外和红外吸收峰,分子质量为1.1×105u,由葡萄糖组成。结论分子质量为1.1×105u的LYRP2-1是由葡萄糖组成,α、β两种半缩醛羟基构型并存的吡喃环多糖。 Aim To extract and purify crude polysaccharides LYRP from Arillus Longan and the monosaccharide constitution of LYRP2-1 and molecular weight. Methods LYRP2-1 was isolated by adding EtOH from the ultrasonic extract of Dimocarpus longan Lour, purified by protease-decomposing method and further purified by DEAE and Sephacryl S-300 gel column chromatographies. The molecular weight and monosaccharide constitution of LYRP2-1 were respectively determined by HPGPC and complete acid hydrolysis. The infromation on the structure of LYRP2-1 was analysed by FT-IR and UV spectroscopy. Results FT-IR and UV spectroscopy of LYRP2-1 revealed typical characteristics of polysaecharides. The monosaccharide constitution of LYRP2-1 whose molecular weight was 1.1 ×10^5 was determined to be glucose. Conclusion The molecular weight of LYRP2-1 is 1.1 ×10^5. Pyranose is the major component of LYRP2-1, which is composed of glucose, with two glycoside configurations, α-hemiacetal hydroxyl and β-hemiacetal hydroxyl coexisting.
