Expression of low-density lipoprotein receptors and hydroxy methylglutaryl-coenzyme A-reductase in human pterygium

摘要 目的:研究LDL-R基因和HMG-CoA-R基因在翼状胬肉组织中的表达。方法:随机选取原发性和复发性翼状胬肉组织各10例,正常结膜组织10眼。实时荧光定量PCR技术分析结膜中HMG-CoA-R和LDL-R的相对mRNA含量。结果:原发性和复发性翼状胬肉中的HMG-CoA-R及LDL-R的mRNA水平均明显高于正常球结膜组织,差异具有统计学意义(P<0.05)。结论:HMG-CoA-R和LDL-R基因可能参与翼状胬肉的发病过程。 AIM: To quantitatively detect gene expression level of low-density lipoprotein receptors (LDL-R) and hydroxy methylglutaryl-coenzyme A-reductase (HMG-CoA-R) in human primary pterygium, reccurent pterygium and conjunctiva. METHODS: Primary pterygium, recurrent pterygium and normal conjunctiva samples were obtained from 30 eyes at the time of surgery during February 2007 to February 2008. Quantitative real-time PCR was used to analyze the content of HMG-CoA-R and LDL-R gene in pterygium and normal tissue. RESULTS: QRT-PCR revealed a statistically significant increase of LDL-R mRNA levels. 4. 1 fold in primary pterygia and 2.7 fold in recurrent pterygium, compared with the control normal conjunctiva. The differences had statistical significance ( P 〈 0.05). Also the mRNA levels of HMG-CoA-R were increased significantly in both primary and recurrent pterygia by 4. 2 fold and 3. 6 fold respectively, compared with normal conjunctiva, with statistical significance ( P〈 0.05). CONCLUSION. This study indicates that pterygium have an altered metabolism of cholesterol namely increased LDL-R and HMG-CoA-R mRNAs, which is characteristic of tumorlike tissues. Both genes may play important roles in the pathogenesis of pterygium.
