摘要
采用叶片针刺接种法从细胞学方面分析转hpa1xoo基因棉花与棉花黄萎病菌互作产生的微过敏抗病反应。通过细胞显微观察表明,转hpa1xoo基因棉花T-34与非转基因棉花在抗病性细胞表型方面存在明显差异。Trypan Blue染色后,伤口外的叶组织中均有一褐色环形条带,褐色环形条带与伤口坏死部位之间区域的组织细胞出现质壁分离和空腔细胞现象,褐色条带与伤口坏死部位之间的区域在转基因和非转基因棉花之间具有较明显的差异,转基因棉T-34较非转基因棉花品种Z35形成的区域小,说明转基因棉细胞损伤程度较非转基因棉细胞损伤程度低。棉花黄萎病菌侵染叶片后,非转基因棉花叶部伤口周围细胞无微过敏性反应(micro HR),受病原菌侵染的叶片萎蔫程度较重;转hpa1xoo基因棉花叶部伤口周围细胞有微过敏反应,叶部刺伤处仅有较小的坏死斑,且不相连,发病较轻,而清水处理的转基因棉花与非转基因棉花叶片中伤口周围均无微过敏反应,说明病原菌侵染诱导转hpa1xoo基因棉花产生微过敏防御反应,转hpa1xoo基因棉花较非转基因棉花有较强的抗病性。
Micro HR(hypersensitive reaction ) produced by the interaction between transgenic hpa 1 xoo cotton T - 34 and Verticillium dahliae Kleb. was studied by via pin - puncturing the leaves. The result showed that cell phenotype of disease resistance in leaves have obvious difference between transgenic cotton T - 34 and non - transgenic cotton Z35. The region between the circular brown band generated by cell Hpid exosmosis and the band of the wound in the non- transgenic cotton Z35's leaves, is wider than that of the transgenic cotton T - 34' s leaves, and no micro - HR was found in Z35, the intensity of leaves wilting is much serious, by contrast, micro HR was fomld around the wound region within the transgenic cotton leaves inoculated with the pathogen, but the micro - HR was not found in the transgenie cotton treated with water and around wound region of non- transgenic cotton Z35 leaves which showed that there was micro- HR defence in transgenic hpa 1xoo cotton. The transgenic hpal xoo cotton had higher resistance reaction to Vertw-dfium dahliae Kleb. than the non-transgenic cotton Z35.
