不连续亲和电泳测定糖化血红蛋白及其临床应用 被引量:2

The Determination of Glycosylated Hemoglobin by Discontinuous Affinity Electrophoresis and its Clinical Application

摘要 利用血红蛋白吸管在醋酸纤维膜上划线加样,不连续亲和电泳的方法分离糖化血红蛋白,并直接洗膜比色。本法去掉染色步骤,减少了检测时间和仪器设备。同时用琼脂法进行盲法检测对照,并比较该法的可靠性。两法同时测定32名健康人及34名糖尿病人,GHb分别为(751±102)%及(72±12)%,(1624±334)%及(1542±24)%。两法结果经相关处理,其相关系数γ=0810;两法无显著差异(P>005);两组GHb测定结果有非常显著差异(P<0001)。该检测方法有利于临床医生对糖尿病人近期血糖变化观察,追踪以及胰岛素用量调整。经追踪糖尿病人的首次GHb,发现凡GHb>125%者,其病情进展变化可能性大,合并症多,必须严格对该组病人血糖给予控制。建议糖尿病人每2~3个月复查一次GHb。 A new detective method of glycosylated hemoglobin (GHb) by discontinuous affinity electrophoresis (DAE) was established in our laboratory.Using DAE method,the separated GHb need not have staining procedure,the Hb washed from cellulose acetate film could be directly determined with colorimetric analysis. The contents of GHb determined by DAE method in 34 patients with NIDDM and 32 healthy controls were(16 24±3 34)% and(7 5±1 02)% respectively.In the same samples,GHb simultaneously determined by conventional agarose gel electrophoresis (AGE) method,their value were(15 12±2 4)% and(7 2±1 2)% respectively,good correlation coefficient of two methods was found (γ=0 810).The results suggest that the value of GHb determined by two methods is very similar (P>0 05),and the DAE method possesses some advantages,such as simple、rapid and suitable for clinical application.
