摘要
目的:克隆及重组苦瓜蛋白MAP30,观察MAP30对大肠癌LoVo细胞凋亡的影响。方法:RT-PCR法克隆MAP30基因的cDNA序列,并表达、纯化MAP30蛋白;MTT法测定纯化的MAP30蛋白对LoVo细胞的生长抑制作用;彗星实验观察MAP30引起凋亡的LoVo细胞基因组的降解情况。结果:成功克隆了苦瓜蛋白MAP30基因;MAP30蛋白对LoVo细胞体外生长具有抑制作用;MAP30蛋白对LoVo细胞半数抑制质量浓度(IC50)为70.0mg/L。MAP30在体外能诱导LoVo细胞凋亡。结论:重组的苦瓜MAP30蛋白在体外能抑制LoVo细胞生长,并能诱导其凋亡。
Aim : To clone and study the inhibitory effect of recombinant MAP30 ( momordica anti-HIV protein of 30 ku) on LoVo in vitro. Methods:Evaluation of drug eytotoxicity to LoVo cells with MTT assay and the obvious apoptotic nuclei of the tumor cells were observed at the appropriate concentrations of MAP30 and the degradation of the genome of the apoptotic cells induced by MAP30 were also dectected by using the method of single cell gel electrophoresis (comet assay). Results : MAP30 possessed very high ability to inhibit the growth of LoVo. The antitumor activities (ICso) to LoVo cells of MAP30 was 70.0 mg/L,and can induce the apoptosis of LoVo cells. Conclusion : The study indicates that MAP30 can inhibit LoVo growth and induce the apoptosis of LoVo cells in vitro.
