摘要
目的探讨雌激素与其拮抗剂他莫昔芬对人乳腺癌细胞系MCF-7高迁移率族蛋白(HMGB1)表达的影响。方法在培养的MCF-7细胞中分别加入无水乙醇、不同浓度的17-beta-雌二醇(E2)和他莫昔芬培养4 d后。用实时荧光定量PCR和Western blot技术检测HMGB1mRNA和蛋白水平。结果17-beta-E2浓度在10-6和10-4mol/L时,HMGB1mRNA和蛋白的表达均显著低于对照组(P<0.05)。他莫昔芬浓度在10-6和10-4mol/L时,HMGB1mRNA和蛋白的表达显著高于对照组(P<0.05)。结论雌激素能够下调HMGB1的表达,而其拮抗剂他莫昔芬则相反。
Objective To investigate the effects of estrogen and tamoxifen on the expression of high mobility group box 1 (HMGB1) in human breast cancer cell line MCF-7. Methods MCF-7 cells were incubated with 17-beta E2 and tamoxifen at different concentrations for 4 days, respectively, with ethanol as control. Real time RT-PCR and Westem blotting were used to detect the expression of HMGB1 at mRNA and protein level after different treatment of the cell line, respectively. Results Compared with ethanol control, HMGB1 mRNA and protein level were significantly decreased when 17-beta-E2 was added at concentrations of 10^-6 and 10^-4 mol/L(P 〈0. 05). And HMGB1 ex- pression was significantly increased when tamoxifen was added at concentrations of 10^-6 and 10^-4 mol/L (P 〈 0. 05). Conclusion Estrogen can down-regulate the expression of HMGB1, while its antagonist tamoxifen has an opposite effect. So, HMGB1 probably has an inhibitory effect on breast neoplasms proliferation.
